ObjectiveThis experiment plan to intervene the epithelial cells of benign prostatic hyperplasia by recombinant human interleukin-6,then detect the apoptosis of cells and the expression of related proteins. In order to investigate the effect of interlenkin 6 on apoptosis of benign prostatic hyperplasia epithelial cell.MethodsSubculture of the BPH-1 cells were divided into groups randomly. One group, respectively, to different concentrations of recombinant human IL-6 (0ng/ml, 2ng/ml, 20ng/ml, 200ng/ml) intervention 48h hours. In addition to another group of IL-6(20ng/ml), respectively,0,4,12,24,48,hour intervention,and then were treated with acridine orange/ethidium bromide staining, ELISA apoptosis detection assay apoptosis in each group and the use of Western Blot assay Caspase-3 activation, Bcl-2 and Bax protein expression; to investigate the IL-6 on prostate hyperplasia epithelial cells apoptosis.Results1,Acridine orange/ethidium bromide staining found in the control group (IL-6, 0ng/ml) can see a large number of apoptotic cells, condensed nuclei yellow, cleavage nucleus; the intervention group with IL-6 (2ng/ml and 20ng/ml) there are rare apoptotic cells; in the 200ng/ml IL-6 group had no obvious apoptotic cells.2,ELISA detection of cell apoptosis find that interfere with the higher concentration of IL-6, ELISA detection of apoptosis in lower optical density, with more IL-6 intervention time (in the same concentration) , intervention and control groups to reduce the percentage of apoptosis.3,Western Blot analysis showed that the intervention group increased with the concentration, Bcl-2 protein expression increased (P<0.05), Bax protein expression was inhibited (P<0.05), Caspase-3 activation was inhibited (P<0.05). Conclusions1,IL-6 can inhibit apoptosis in BPH-1 cells, and was time and dose-dependent, IL-6 concentration levels higher, the BPH-1 cell apoptosis in the stronger inhibitory effect, the longer time the intervention of BPH -1 the stronger the inhibition of apoptosis.2,IL-6 can promote the Bcl-2 protein expression of the BPH-1 cells inhibited Bax protein expression and inhibit the activation of Caspase-3.
