Objective Probing into the Traditional Chinese Medical Pathologensis of benign prostatic hyperplasia .To study the relationship of cell signal conduction and benign prostatic hyperplasia, clarify the mechanism of Xian Jia prescription restraining benign prostatic hyperplasia, and reveal the asthenia of kidney and blood stasis' scientific content of benign prostatic hyperplasia.Methods (1) S. D male rats were divided into 6 groups : normal group, model group,Long Bi Shu group, Xian Jia prescription low dose group, middle dose group and high dose group.Rats were castrated ,then subcutaneous injected testosterone propionate for 30 days. (2)Observe the changes of prostate weight , prostate exponent, histology under light microscope of each group. (3)Observe the changes of ultrastructure under electon microscope. (4)Take the radioimmunochemistry to measure the sex hormone content of prostate constitution. (5)Have in situ hybridization to detect the expression of PKAmRNA and PKCmRNA. (6)Use the method of immunohistochemistry to detect the expression of EGF, TGF-B1, bcl-2, bax in each prostate constitution. (7)Take the Flow Cytometry to measure the apoptosis rate of each group.Results (1)The protate weight, epithelia cell height increased significantly (P<0. 01) ,Xian Jia prescription can restaint the changes. (2)Model rats have obvious anomaly in histology and ultrastructure in prostate, Xian Jia prescription can relieve the anomaly. (3)The contents of dihydrotestosterone (DHT) in model rats increased greatly (P<0. 01) , Xian Jia prescription can low the level of DHT. (4)The expression of PKCmRNA in model prostate cells increased significantly,but the PKAmRNA decreased greatly, Xian Jia prescription can adjust PKCmRNA/ PKAmRNA. (5)The expression of EGF,bcl-2 in model prostate cells increased greatly, but the TGF-B1, bax decreased significantly, Xian Jia prescription can regulate EGF/TGF-B1, bcl-2/bax. (6)The apoptosis rate of prostate cell in model rats decreased significantly (P<0. 01) , Xian Jia prescription can increased the apoptosis rate of prostate cell.Conclusion The study has suggested that the asthenia of kidney and blood stasis is the fundmental pathogenesis of benign prostatic hyperplasia. By the experimental study ,we detected the DHT's content aggregated,the cell apoptosis rate decreased,the expression TGF-B1, bax ,PKAmRNA reduced and EGF,bcl-2 ,PKCmRNA increased in model group. The results suggest that DHT aggregation,growth factors, protein kinase and apoptosis gene's disorder ,lead to the apoptosis rate decreasement, result in benign prostatic hyperplasia. Xian Jia prescription can significantly control weight of rats' prostate ,wepresume Xian Jia prescription can regulate the signal conduction of sex hormone, growth factors and protein kinase .effect on apoptosis gene , induce the cell apoptosis of benign prostatic hyperplasia. EGF/TGF-B1, bcl-2/bax and PKCmRNA/ PKAmRNA incline to normal, benign prostatic hyperplasia was restrained.
