Objective①To observe disposition of proliferating cell ki-67 nuclear antigen in architecture of prostate cancer(PCa),benign prostate hyperplasia(BPH) and normal prostate(NP).②To observe disposition of the related apoptosis genes proteinum of bcl-2 and bax.③To observe disposition of anti-oncogene p53 in architecture of prostate caner,benign prostate hyperplasia and normal prostate.④To observe relationship between prostate-specific antigen(PSA) and the expression of the related genes of ki-67,bcl-2,bax and p53.⑤To observe regularity of disposition of ki-67,bcl-2,bax and p53 in architecture of prostate gland,observe interaction and relationship during morbility of prostate cancer,benign prostate hyperplasia.⑥Through deep approach about these relationship in order to deep comprehend etiopathogenisis of PCa and BPH,in order to offer assistance for theoretical investigation and clinical iateria and clinical diagnosis.Methods Collect the 36 cases of exairesis specimen of PCa,20 BPH and simul collect the 11 cases of exairesis specimen of NP,to progress research about cell multiplication and apoptosis:①Hematoxylin-Eosin(HE) staining.②The expression of ki-67 protein and bcl-2,bax and p53 protein were detected by immunohistochemical S-P method reflected proliferating index(PI) with genetic ki-67 nuclear antigen and apoptosis with apoptosis genetic bcl-2,bax and anti-oncogene p53.③PSA of serum in determine prostate-specific antigen with radio-immunity method.④The computer image analysis.⑤Disposed the data of the computer image analysis by SPSS for Windows12.0 statistical software. Results①Observation of HE staining:Normal prostatic is made of glandular organ and ground substance.It is thus clear that glandular tissue and ground substance tissue is hyperplasy in benign prostate hyperplasia.It is divided 4 stages of cell in prostate caner according to differentiated degree of glandular organ and more nature of cell and dyskaryosis on the basis of Gleason grading standard.②Immunohistochemistry:The active number gradually increase of ki-67 proteinum in NP,BPH,PCa.PI expressed obviously higher of BPH than that of NP(P<0.01),PI expressed obviously higher of PCa than that of BPH also(P<0.01),ki-67 expression correlated with pathologic grading of cancer,increased as PCa increases in grades,its dependablity is obviously positive correlation(r=0.248,P<0.01),bcl-2 positive expressing rate was 30%and 44% in PCa and BPH,higher than that of NP 9%(P<0.05),bcl-2 expression correlated with pathologic grading of cancer,increased as PCa increases in grades,were respectively 17.3%and 46.2%,its dependablity is obviously positive correlation(r=0.375,P<0.01), bax positive expressing rate difference had no significance in PCa,BPH and NP,were respectively 100%,85%,80.6%(P>0.05).The active number gradually increase of p53 proteinum in NP,BPH and PCa.Its positive expressing rate were respectively 0%,5%and33.3%,expressed obviously higher of PCa than that of BPH also(P<0.01). p53 expression correlated with pathologic grading of cancer,increased as PCa increases in grades,was respectively 21.7%and 61.5%,according to statistical treatment.its dependablity is obviously positive correlation(r=0.489,P<0.05).③Prostate-specific antigen's concentration of blood in PCa was 35.6±18.53μg /ml.Its concentration is obviously positive correlation with ki-67,bcl-2 positive expressing rate(P<0.05). Prostate-specific antigen's concentration of blood in BPH was 5.85±3.761μg /ml.Its concentration was not obviously correlation with ki-67,bcl-2 positive expressing rate(P>0.05).Conclusions①Comprehend morphologically variant distinction of tissue in prostate gland from observation of HE staining of morphologically prostate gland.②Disposition of cell proliferation related gene ki-67 had regularity:ki-67 expressed obviously higher of PCa than that of BPH and NP.Expression of ki-67 correlated with pathological grade, increased as PCa increases in grades.Its dependablity is obviously positive correlation.③Positive expressing rate of bcl-2 was higher of BPH and PCa than that of NP.bcl-2 expression correlated with pathological grade,increased as PCa increases in grades.Difference of positive expressing rate of bax had no significance in.PCa,BPH and NP.④Positive expressing rate of p53 was higher of BPH and PCa than that of NP. p53 expression correlated with pathological grade,increased as PCa increases in grades.Its dependablity is obviously positive correlation.⑤Prostate-specific antigen's concentration of blood in PCa correlated with pathological grade,increased as PCa increases in grades.It is obviously positive correlation with positive expressing rate of ki-67 and bcl-2.⑥Cell proliferation and apoptosis related gene ki-67,bcl-2,bax and p53 play an important role in development of BPH and PCa.so it possibly lead to formation of BPH and PCa.⑦From aggregate analysis of cell proliferation and apoptosis,it was furthermore sense to evaluate genesis,development and prognosis of BPH and PCa.It will provid a new thinking way for clinical therapeutics and iateriaclinical diagnosis of BPH and PCa.Its effect is possibly furthermore ideal of protein binding of ki-67,bcl-2 and PSA of blood for diagnosis of PCa.
