Objective:To discuss the underlying mechanisms of lacidipine to defend Hcy-induced endothelial cell damage through viewing effects of lacidipine to antagonize inhibition of HUVEC proliferation caused by Hcy,inhibition of formation of TLS and the effects on the expression of VEGF.Methods:After isolate-culturing HUVEC,the opposition effects of lacidipine on proliferation inhibition of HUVEC caused by Hcy were measured by MTT way at different concentration;The changes of cell cycle of the cultures after administration of lacidipine and Homocysteine were observed using flow cytometer;the influence of lacidipine on the HUVEC tubulus-like structure formation was measured in TUBE canaliculization formation experiment;the effects of lacidipine on the expression of VEGF were determined by real-time-PCR.Results:lacidipine could defdefat proliferation inhibition of HUVEC induced by Hcy at 1*10-3 mM,1*10-2 mM,which showed a signicant difference compared with Hcy-induced damage group(P<0.05)but had no difference VS blank-control group(P>0.05),and lacidipine had no effects on Hcy-abscent cultured HUVEC proliferation VS blank-control group(P>0.05),while all above-metioned effects did not exist at 1*10-4 mM.In TUBE canaliculization formation experiment, tubulus formation decreased in Hcy group,but lacidipine would antagonize this effect at 1*10-2 mM,1*10-3 mM while had no influence on tubulus production in Hcy-abscent group.Lacidipine could obviously enhance VEFG expression in Hcy-damage group at 1*10-2 mM,1*10-3 mM measured by realtime-PCR,which showed a signicant difference compared with damage model group(P<0.05),but had no difference VS blank-control grgroup(P>0.05).Conclusions:lacidipine could defend Hcy-induced inhibition of HUVEC proliferation and inhibition of formation of TLS.The underlying mechanism may relate to the up-regulation of VEGF expression,which hint that lacidipine may play a role in the genesis and development of new vessels of CAD ischemic cardiac muscles.
