ObjectivesThis research observed the express of nestin protein in neural stem cells and the express of NSE protein in neuron cells of SD rats treated by angelica and sodium ferulate injection, in order to find weather injections mentioned above could promote neuron regeneration.MethodsThirty-three female rats were divided into 3 groups: the angelica injection group, the sodium ferulate injection group and the control group. Rats' models of spinal cord hemisection were made and the angelica and sodium ferulate group and controls were given respectively the 25% angelica injection and sodium ferulate injection by continuously intraperitoneal injection while physiological saline was injected into abdominal cavity for 3 weeks after injury. Then the rats were sacrificed and the 5mm length of cord segment around lesion is cut down for test. The recovery of the locomotor function after the injury was evaluated with Basso, Beattie and Bresnahan (BBB) scale, and degree and scope of the spinal cord regeneration after injury were assessed using Immuno- histochemistry to observe Nestin and NSE expression in leisons of the spinal cord.ResultsThe number of nestin and NSE positive expression cells and the region of nestin and NSE positive expression dominated in the site of injury and the adjacent tissues of the angelica injection group is more than that of the sodium ferulate injection group, which is more than the control group, existing significant level among three groups. BBB scores show no significant level among three groups.Conclusion1. The angelica and sodium ferulate injection can promote the activation and conglomerat of neural stem cells and induce the nerves regeneration after cord injury in rads, but not having a effect of locomotor fuctional improvement. 2. There exists significance level between effects treated by angelica and sodium ferulate injection on endogenous neural stem cells after acute spinal cord injury (SCI) in rats, that means there is some effective ingredients else which could promote the recovery of the locomotor function after the injury. If we can find the mechanism in molecules and pharmacology and enlarge the treating effects, we get more close to clinical spine recovery.
