Objective:To investigate the effect of meglumine cyclic adenylate (MCA) on endogenousneural stem cells after spinal cord injury (SCI), then discuss the possible mechanism.Methods:Ninety adult SD rats were randomly divided into3groups: The sham operation group(group A, n=30), control group (group B, n=30) and MCA treatment group (group C, n=30).SCI was simulated at T11level by Allen’s method both in group B and C, but group A wasgiven vertebral lamia fenestration only,but not be damaged.The rats in group C were injectedwith MCA2mg/kg/d after SCI, until draw materials, while the animals in group B receivedsame amount of0.9%normal saline likewise. The function of hindlimb movements wereevaluated by BBB scales and as a reference to assess the repairing effect of treating strategy,and were taken specimens for HE staining at1d,3d,5d,1、2and4weeks after injuryrespectively, and then the radius within1.0cm of the injury center were analyzed by Nestinimmunoreactivity.Result: Group A rats hindlimb movements function all normal at different time points, group Band C movements function scores were all lower than the normal. But the intervention grouprats the hindlimb function were more better than control group after five days (P<0.05).Therewas little Nestin expression in ependymal cells and seldom observed in white matter in group A.In group B, Nestin expression was observed obviously in ependymal cells near the central canalat1day after injury, which reached peak at1week time point (P<0.05). Moreover, Nestinexpression was also observed migrating from the central canal toward the gray matter. Theexpression decreased in ependyma cells at2weeks time point, and expressed a few at4weeks.At1day time point, Nestin expression in group C was no different from group B. But thedifferences appeared from3days time point. At1week, its positive expression was more thangroup B, especially surround the central canal (P<0.05), and high expression still existed at2weeks,which last to4weeks.Conclusion: MCA can promote endogenous neural stem cells proliferation after acute SCI inrats, and induce polarization to neurons. And it also showed that MCA can be used to promotingthe repair of nerve damage after acute spinal cord injury.
