Objective:To observe the effects of Jiaweibugan decoction on expression of BDNF protein and BDNF mRNA in sciatic nerve of experimental diabetic rats in order to explore the preventive and therapeutic effects of Jiaweibugan decoction on peripheral neurophathy in experimental diabetic rats.Methods:Randomly selected 20 rats form 80 male Wistar rats as normal contral group, and remnant 60 rats used by model building. The diabetic neuropathy rats model was reproduced by streptozotocin. The veil blood was collected from vein to measure blood glucose after 72 hours, successful diabetes model rats whose blood glucose is greater than 16.7mmol/L. The 60 rats were randomly divided into three groups including model-contral groups; a cobalt ammonium groups and Jiaweibugan decoction-treated group. Immunohistochemical method was used to test the level of protein of BDNF in sciatic nerve and RT-PCR was used to determine the expression of BDNF mRNA in sciatic nerve in 4th and 8th weekends from the beginning of treatment respectively.Results:(1) The effects of Jiaweibugan decoction on blood glucose and weightxompare with the model group, Jiaweibugan decoction and a cobalt ammonium have no significantly ameliorating to weight and blood glucose of the rats with diabetes induced by streptozotocin(P>0.05).(2) The effects of Jiaweibugan decoction on the sciatic nerve conduction velocity: The velocity in model group decoction was slower than that in normal control group(P<0.05), and that in a cobalt ammonium and Jiaweibugan decoction was faster than that in model group(P<0.05), but there was no signifiacant difference between a cobalt ammonium and Jiaweibugan decoction.(3) The effects of Jiaweibugan decoction on the BDNF protein and BDNF mRNA expression in sciatic nerve: The expressions of BDNF protein and BDNF mRNA in sciatic nerve of model rats were lower than those in the normal control group(P<0.05), while those 8 weekend were higher than in those 4 weekend in model rats. while those in model rats were treated by Jiaweibugan decoction and a cobalt ammonium were markedly higher than in those non-treated model rats(P<0.05), but there was no signifiacant difference between a cobalt ammonium and Jiaweibugan decoction(P>0.05).Conclusions:(1) In high glucose conditions, the expressions of BDNF in sciatic nerve decreased, weaked the repair capacity that Damaged nerve fibers. This may be one of the reason of damages to structure and function of peripheral nerves.(2) Jiaweibugan Decoction can increase the expressions of BDNF in sciatic nerve of diabetic rats, aecoction can increase the expressions of BDNF in sciatic nerve of diabetic rats. This may be one of the function mechanisms of protective effects of Jiaweibugan Decoction on diabetic peripheral neuropathy.
