Implication Of RAPD In Identification Of The Merchandise Drug Polygonatum Odoratum

Yuzhu is the rhizome of Polygonatum odoratum(Mill.) Druce(Fam. Liliaceae),which is not only a common Chinese drug but also a genuine drug in Hunan.Many research methods,such as morphological identification,anatomical identification and physical and chemical identification,have been applied in the study on Yuzhu.My staff room had reported the molecular evidence in identification of Yuzhu firstly. This work is based on original work.The technique of random amplified polymorphic DNA(RAPD),as a DNA molecular index,was used in the study which included not only merchandise drug Yuzhu from different habitats in China and familiar drug huangjing.This study probed into the implication of molecular technique in identification merchandise drug yuzhu and familiar drug huangjing,discussed genuine drug,established a new methed to identification varieties of merchandise drug Yuzhu.This study not only maked up shortage but also study on inheritance furthermore. In this study,the establishment of the RAPD fingerprinting of merchandise drug Yuzhu was performed,which included the optimised ingredients of the RAPD and selected the random primers.The data based on the RAPD were used to generate Nei similarity coefficients and to construct a tree diagram using the UPGMA in the NTSYS programs. The results and conclusions:(1) The furthest methed of extraction DNA has been established;(2)The RAPD reaction system suitable for Yuzhu and Huangjing was established;(3) 24 primers were screened from more than 200 random 10-mer primers,and a totle of 162 DNA bands were amplified,143of which(88.3%) were polymorphics;(4) The RAPD results reach the same conclusions as those obtained from the studies on morphology and cytology;(5) There was a rich genetic polymorphism between Yuzhu and Huangjing.(6)It was evident to see the genetic difference between the genuineness of Yuzhu and the different habitats of Yuzhu.This study firstly applied the RAPD technology to identify merchandise drug yuzhu and familiar drug huangjing,discussed difference between Yuzhu in Hunan and other different habitats of Yuzhu.,which was a beneficial try to add a new method into the merchandise Chinese traditional medicine identification.
