The Study Of The Relativity Between The Expression Of MMP-3 And Morphologic Change In Cartilago Acetabularis In DDH

Objective:To determine the expression of matrix metalloproteinase-3 (MMP-3) and morphologic change of cartilago acetabularis in developmental dislocation of the hip(DDH) in order to investigate the relationship between the expression of MMP-3 and dyschondroplasia or cataplasia of carti!ago acetabularis in DDH.Mothods:DDH groups(T group,30 cases cartilago acetabularis) were divided into T₁ group(13 cases,2-5 years) and T₂ group(17 cases,5-14years) and 15 cases of the cartilago acetabularis of the children in control group(C group) were got from the department of the anatomic in Zunyi Medical College.The general observation,the histomorphology of by HE and the expression of MMP-3 by DAB immunohistochemical method were observed in all samples of the cartilago acetabularis.The data of the result was analysed by the SPSS13.0 statistical software.Results:(1)The smooth were observed on the surface of cartilago acetabularis in control group and the flexibility and transparency of the cartilago acetabularis was decreased and the surface of the cartilago acetabularis was rougher and irregularity in DDH group(T group).(2)The homogeneous cartilage matrix was the main part,the symmetrically array cartilage cells were gradually lessen from surface to inside and the cell nuclears were circle and parallel arrayed on the surface of cartilago acetabularis,the nuclears presented ellipse,histo-arrayed at inside in control group.Inhomogeneous and thin of staining-phil of cartilage matrix,fibrosis and turbulence of arrangement of fasciculi were observed and the arrangement asymmetry and cluster phemonenon of chondrocyte were found,the obviously decrease of chondrocyte,pyknosis of nucleus and the lacune augmentated or diminished were observed in severe cataplasis of cartilage in DDH group(T group).(3)There were brown yellow particles in positive ceils of expression of MMP-3 and in the mainly cytolymph and 17 cases(56.7%) of positive in T group, one positive(6.67%) in control group.The positive rate of the expression of MMP-3 in T group was lfigher than that in C group(p＜0.05),there were 2 weak positive,3 positive and no strong positive(16.67%) in T₁ group and 5 weak positive,6 positive and 1 strong positive(40%).The positive rate of the expression of MMP-3 in T₂ group was higher than that in T₁ group(p＜0.05).Conclusions:1.The positive rate of the expression of MMP-3 in cartilago acetabularis in DDH group(T group) is higher(56.67%) than that in C group(6.67%) and there is significant statisticsing meaning;2.The positive rate of the expression of MMP-3 in T₂(40%) is higher than that in T₁(16.67%) and there is a positive correlation between the expression of MMP-3 and age and the expression of MMP-3 increased while the age increased;3.The developed unwell and serious degeneration of cartilago acetabularis cells are found in DDH group(T group) with general observation as well as the result of HE,the positive rate of expression of MMP-3 and morphologic change are the correlation,MMP-3 could be one of the important causes in pathological development of DDH.
