Object:To apply micronucleus test combine the chromosome metaphase analysis and gene detection to analyze preliminary diagnosis leukemia patient's genetic damage,and the relationship between the results of Micronucleus test and prognosis correlation factor.Preliminary investigate the value of micronucleus test in leukemia nosogenesis,diagnosis,prognosis study.Methods:To apply Cytokinesis-blocked micronucleus test analysis 54 preliminary diagnosis leukemia patient's bone marrow and peripheral blood compare the MNR,MCR,Buds,NPBs,NDI,AC with 30 healthy people's peripheral blood's results. (1)compare the MNR,MCR between the leukemia patient's bone marrow and peripheral blood,analysis the different;(2)combined the chromosome metaphase analysis and gene detection as the target of the chromosome damage,compare with healthy people's,to analysis the leukemia patient's inheritance damage;(3)select 31 complete case,combined the sex,age,the count of the white blood cell,the count of the hemoglobin,the count of the platelet,the proportion of the naive cell in bone marrow,chromosome metaphase analysis result,gene detection,analysis the correlation with the result of the micronucleus test.Results:1.The 54 preliminary diagnosis leukemia patient's peripheral blood's MNR is 11.63±3.05‰,MCR is 10.81±4.92‰;bone marrow's MNR is 12.42±1.62‰, MCR is 11.13±3.75‰,though the t test,P value>0.05,no statistical significance differences.2.The 54 preliminary diagnosis leukemia patient's peripheral blood's MCR is 10.81±4.92‰,30 healthy people's peripheral blood's MCR is 3.81±2.21‰, though the t test,P value<0.01,existence statistical significance differences. The leukemia patient's peripheral blood's MNR is 11.63±3.05‰,healthy people's peripheral blood's MNR is 3.81±2.21‰,though the t test,P value<0.01,existence statistical significance differences.The leukemia patient's peripheral blood's Buds 3.50±2.93%,healthy people's peripheral blood's Buds 0.20%±0.40,though the x~2 test,P value<0.05;existence statistical significance differences.The leukemia patient's peripheral blood's NPBs is 1.88±1.89%,healthy people's peripheral blood's NPBs is 0.40±0.80%,though the x~2 test,P value<0.05 existence statistical significance differences.The leukemia patient's peripheral blood's NDI is 1.51±0.30%,healthy people's peripheral blood's NDI is 1.22±0.32%,though the x~2 test,,P value>0.05, no statistical significance differences.The leukemia patient's peripheral blood's AC is 2.23±1.33%,healthy people's peripheral blood's AC is 0.16±0.37%,though the x~2 test,P value<0.05;existence statistical significance differences.The leukemia patient's the abnormality ploid detection rate though chromosome metaphase analysis is 24.00%,healthy people's the abnormality ploid detection rate is 0.00%,though the x~2 test,P value<0.01;existence statistical significance differences.The leukemia patient's fusion gene rate is 18.00%,healthy people's fusion gene rate is 0.00%,though the x~2 test,P value<0.01;existence statistical significance differences.3.Though the correlation test,the MNR,MCR have the correlation with age and the abnormality chromosome ploid,the NDI have the correlation with the count of the white blood cell.Conclusion:1.The preliminary diagnosis leukemia patient's MNR,MCR,Buds,NPBs,AC,the abnormality detection rate though chromosome metaphase analysis,fusion gene rate all higher than healthy people's corresponding results.The leukemia patient has inheritance damage in the initial stage.2.The preliminary diagnosis leukemia patient's bone marrow's MNR and MCR a litter higher than peripheral blood's,but have no statistical significance.Both bone marrow and peripheral blood can be the material of the micronucleus test.3.The value of results of micronucleus test about preliminary diagnosis leukemia patient as both the value of leukemia patient prognosis factors still need further research.
