Objective:Contracting the effect of peripheral endothelial progenitor cells which has been drived by Simvastatin in different path and way,viewing the amount of peripheral endothelial progenitor cells at 6 time points,comparing the fit and unfit by the two ways of administration,To provide some new ideas and simple method for EPCs(endothelial progenitor cells) transplantated and other artificial stent to vascularize rapidly,as well as provide a reference for the tissue-engineering skin to vascularize.Methods:To choose raw material of Simvastatin(LuNan clique,ShanDong province,production number:050502) that has been used in clinical,to mobilizate by the dose in accordance with the literature.Selected healthy mouse(to avoid the effects of estrogen on EPCs) ICR mouse 270,randomly divided into 3 groups:A group (Simvastatin intraperitoneal injection),B group(Simvastatin intragastric administration),C group(negative control),90 in A,B group,30 in C group.And A group(Simvastatin intraperitoneal injection) is provided into three subset groups:A1 group(Simvastatin with high does:15mg/Kg*d-1),A2 group(Simvastatin with low does:2mg/Kg*d-1),A3 group(normal saline control),And B group(Simvastatin intragastric administration) is also provided into three subset groups:B1 group (Simvastatin with high does:15mg/Kg*d-1),B2 group(Simvastatin with low does:2mg/Kg*d-1),B3 group(normal saline control),in each subset grou there are 5 mouse to select randomly.And each subset group is designed with 1d,3d,5d,7d,10d,14d six time points,at witch the mouse are taked blood for Flow cytometry CD34,Flk-1,CD133-positive labeled cells.The results were used(?)±s to express, were analyzed by statistics.Results:The mount of peripheral endothelial progenitor cells in contracted subset groups of A group(Simvastatin intraperitoneal injection) and B group (Simvastatin intragastric administration) has been no significant difference at 6 time points(p>0.05);There is no significant difference between normal saline control group and negative control group at 6 time points(p>0.05);the group of Simvastatin with high does has been gradually descended at 1d,3d,5d,7d time point,7d is the lowest,10d,14d retain in this level(p<0.05),There is significant difference between high does group and saline control group at 6 time points(p<0.05);the group of Simvastatin with low does has been gradually ascensused at 1d,3d,5d,time point,5d is the lowest,7d,10d,14d retain in this level(p<0.01),There is significant difference between low does group and saline control group at 6 time points(p<0.01).Conclusion:the quantity of EPCs in peripheral blood can,be increased by Simvastatin with low does mobilizating;on the contract,the quantity of EPCs in peripheral blood can be inhibited by Simvastatin with high does mobilizating;the two ways of Simvastatin intraperitoneal injection and Simvastatin intragastric administration has the same effect in mobilizating EPCs.
