Objective To investigate the expression levels and significance of transmembrane protein endothelial factor(TPEF) , signal transducer and activaon of transcription 1 (STAT1) and cyclin-dependent kinase 8 (CDK8) in colorectal cancer and their correlations with clinicopathologic parameters.Methods Expression levels of TPEF mRNA , STAT1 mRNA, CDK8mRNA were detected by SYBR Green I quantitative real-time PCR in colorectal cancer and corresponding normal mucosa in 50 patients; analysis was performed to study on the correlation between the expression levels of TPEF mRNA , STAT1 mRNA, CDK8mRNA and clinicopathological parameters, and on the relationship between expression level of TPEF with STAT1.Results Expression levels of TPEF mRNA and STAT1 mRNA in colocrectal cancer [0.2790(0.0687,0.8237),1.2005(0.5340,2.2645)] were significantly decreased than that in normal mucosa [0.8020(0.3072,1.9467),1.6835(0.7470,2.7192)] (P<0.05). Expression levels of CDK8 mRNA in colocrectal cancer [1.1055(0.6262,1.7770)] were significantly increased than that in normal mucosa [0.5545(0.2910,1.0022)] (P<0.05). The expression levels of TPEF mRNA and STAT1 mRNA in non-lymph node invasive group [0.7155(0.0757,1.5505),1.8435 (1.2072,2.4717)] were significantly higher than that of the lymph node invasive group [0.1150(0.0277,0.2790),0.6100(0.3895,1.1590)] (P<0.05). The expression levels of CDK8 mRNA in non-lymph node invasive group [0.8535 (0.3542,1.7072)] were significantly lower than that of the lymph node invasive group [1.1695(0.8740,2.7317)] (P<0.05). The expression levels of TPEF mRNA and STAT1 mRNA were also found more frequently in stage T1/T2 cancer group [0.7550(0.2730,1.3750),2.3420(1.5755,2.9700)] than in stage T3/T4 group [0.1145(0.0367,0.4415),0.9975(0.4172,1.3427)] (P<0.05); The expression levels of CDK8 mRNA were also found more frequently in stage T3/T4 cancer group [1.5915(0.9965,2.2892)] than in stage T1/T2 group [0.5745(0.4005,0.9587)] (P<0.05). There was a positive correlation between the expression level of TPEF mRNA and STAT1 mRNA in colorectal cancer and corresponding normal mucosa(rcancer=0.537, rnormal=0.851,P<0.001).Conclusion1. TPEF, STAT1 and CDK8 correlated with colorectal cancerous tissues, and might be used as predictive indicators for the malignant potential of colorectal cancer.2.
