Qingqiliangying Injection is a traditional Chinese medicine prescription,which is composed of Folium Isatidis,Flos Lonicera,Radix et Rhieoma Rhei,Rhizoma Anemarrhenae,Chrysanthemum indicum and Lophatherum gracile Brongn.. Clinically,it always used to manage the epidemic hemorrhagic fever,angina parotidea and so on.Qingqiliangying Injection has been already studied in 1997 by the Qingqiliangying study team.On basis of the study in 1997,we use macroporous resin isolation technic to separate the active ingredients of Qingqiliangying Injection. And then at the result of the screening of rats' antipyretic efficacy,use HPLC and HPLC-MS to analyze the ingredients of the injection.At last,we expect to find the attribution of the ingredients and determin the antipyretic ingredients of Qingqiliangying Injection.First use the static adsorption and desorption,the dynamic adsorption and desorption to screen the proper resin.Use the solid content of Qingqiliangying Injection as index.The result shows that the resin of HPD-100 is the best one of the tested resins.Use water,30%alcohol and 95%alcohol to elute Qingqiliangying Injection after screening the solvent.And the separation is well.Use the rat febrific model induced by 10%zymonsa solution to observe Qingqiliangying Injection and its separated parts' effect on the rat temperature. Furthermore use the radioimmunoassay(RIA) to determin the content of PGE2 and cAMP in rats' hypothalamus in order to determin the antipyretic effect of the injection.The result shows that Qingqiliangying Injection is quite effective for pyretolysis.Its water separated part doesn't show any antipyretic effect.Its separated part by 30%alcohol and 95%alcohol can reduce the rat temperature.And the antipyretic effect of 30%alcohol separated part is stronger than the 95%alcohol separated part.Use the HPLC to analyze Qingqiliangying Injection,its 30%alcohol separated ingredients and blood-serum contained Qingqiliangying Injection,and then find the attribution of the peaks.We use three mobile phases to analyze the hydrophilia part, lipophilia part and aglycone part,respectively.Deal the six traditional Chinese medicines with the same method as Qingqiliangying Injection.Compare the retention time and UV spectrogram of Qingqiliangying Injection with traditional Chinese medicine's.Initially estimate the attribution of the ingredients.Compare the retention time and UV spectrogram of Qingqiliangying Injection with blood-serum's. Initially estimate the attribution of the ingredients in blood-serum.The result shows that major ingredients of Qingqiliangying Injection belong to Folium Isatidis,Flos Lonicera,Radix et Rhieoma Rhei and Chrysanthemum indicum,and the 30% alcohol separated part belong to Flos Lonicera,Radix et Rhieoma Rhei and Chrysanthemum indicum.Judging from the attribution of the ingredients in blood-serum,we can estimate that the antipyretic ingredients may come from Chrysanthemum indicum.Use HPLC and HPLC-MS date to determine the exact chemcial constitutions of the ingredients in Qingqiliangying Injection and serum.The result shows that 17 chemical compounds detected from Qingqiliangying Injection can be determined. And one of the determined chemical compound in serum attributed to Chrysanthemum indicum is determined as linarin.
