[Objective]To detect the abnormal expressive microRNA in the serum of lung cancer patients,and would develope some novel biomarkers for quickly diagnosis of early lung cancer.This test provides a experimental basis for constructing micorRNA expression profile in the serum of lung cancer patients.[Methods]Serum samples were collected from 4 lung cancer patients without any treatment, and from 2 healthy volunteers.The samples' RNA extract were screened with the miRNA gene chip(Capitalbio Corporation) for the different expression of miRNA.The results will be tested by real-time PCR to choose the obviously disregulation genes for forecasting relative target genes.[Results]1,36 miRNAs target genes up-regulated and 57 miRNAs genes down-regulated in serum from lung cancer patients were detected.Among the miRNAs above,miR-320a/b/c, miR-1225-3p,miR-1228~*,miR-1237,miR-1249 were up-regulated obviously;and miR-10a, miR-30a/d,miR-34b,miR-181a-2~*,miR-192,miR-296-5p,miR-454~*,miR-563,miR-576-3p,mi R-598,miR-653,miR-664,miR-885-3p,miR-1184,miR-1826 were down-regulated obviously. 2,It was confirmed that results of real-time PCR were not consistent with the gene chips.3,The 2 miRNA s up-regulated obviously were choosed to forecast target gene.[Conclusions]1,There are circulating miRNAs in serum of lung cancer patient actually.The miRNA in surm may be a kind of novel biomarkers.2,we chose 23 disregulated miRNAs in serum of lung cancer patients using the miRNA gene chip V3.0(Capitalbio Corporation).3,It was confirmed that results of real-time PCR were not consistent with the gene chips.4, we choose miR-320a and miR-320b which were up-regulated obviously to forecast target gene.The result showed that the 2 miRNAs are related with tumor.
