Objective: Inhibitory effect of Artemether on U251 glioma cells in vitro, to find out that Artemether have inhibitive effect on U251 intracranial glioma growth and angiogenesis in tumor bearing mice,and also to study the possible mechanism of artemether inhibiting tumor,and also to find a new method for the treatment of human gliomas.Method: 1. Culture of U251 cells into vitro, then we use MTT (Methy thiazolyl tetrazolium) to evaluate the inhibitory effect of Artemether treatment on U251 glioma cells. The expression of VEGF of U251 glioma cells treated with Artemether was detected by enzyme-linked immunosorbent assay (ELISA). 2. Establishment of U251 glioma model,stereotaxic localization of anesthetized mice,the first cut the scalp, Location of injection points in the vicinity of the sagittal and coronal sutures,Bone marrow was filled with bone wax and sutured to the head after injection of glioma cells at the site.Observe the survival atatus of mice,Brain tissue was removed and fixed after epilepsy in mice Tumor growth was determined by gross observation and HE.3.To establish an in situ model of U251 glioma,Twenty-four SCID mice were randomly into three groups. They were blank control normal saline group(0.4ml /10g/d) as oral; control group Artemether(50.0mg/kg/d)as oral; positive control group Temozolomide (20mg/ kg/d) ip. Temozolomide was administered by gavage.Every time the medicine and observe the survival atatus of mice,About 20 days,When the mice were epileptic,we took out the brain tissus,Tumors were collected for HE staining and growth status of tumor was compared among each group. The expression of vascular endothelial growth factor (VEGF) in tumor cells in each group was observed by immunohistochemistry and was compared by statistical analysis.Results: 1. Artemether can inhibit the U251 glioma cells growth and inhibit the formation of new blood vessels.And depended on the dose and exposure time.The content of VEGF in the cell liquid of Artemether was decreased obviousy. the expression of VEGF in U251 glioma cells was obviously lower than control group(P<0.05). 2. Mouse U251 glioma model was successfully consturcted, Tumor was found. 3. Compared with control group,artemether could significantly prolong the survival time of mice,(P<0.05); After nuclei of U251 glioma cells in the control group were increased and staining deepened after brain HE, At low magnification,the U251 glioma cells infiltrated into normal brain tissue.Conclusions : 1. Artemether can inhibit the proliferation of U251 glioma cells and the effect was time and dose dependent.2.Artemether may suppress the glioma tumor growth probably by inhibiting Angiogenesis.3.Artemether could inhibit U251 glioma in mice.
