Study Of ICAM And Platele Membrane GPIbα Gene Polymorphisms In Patients With Cerebral Infarction Disease

objectives 1 To explore the correlation between plasma levels of soluble ICAM and the attack of acute cerebral ischemic stroke (ACIS) and the changes of platelet membrane glycoprotein (GP) Iba (CD42b) on peripheral blood in ACIS patients; 2 To learn the distribution frequencies of GPIba genetypes in Kunming Han population; 3 To reveal if the gene polymorphism of GP Iba are associated with the occurrence of ACIS; 4 To reveal if the plasma levels of the CD42b is affected by the gene polymorphism of GP Iba.Methods The expression of CD42b from peripheral blood was measured by flow cytometry and the level of ICAM was measured by enzyme-linked immuno-sorbent assay in 119 case of patients with ACIS and 117 controls. The VNTR polymorphism on platelet membrane GPIba were analysis by Polymerase chain reaction-restriction fragment length polymorphism, DNA sequencing analysis was applied in 20 products which randomly selected.Results 1 Compared to controls, the level of sICAM-1, sICAM-3 was increased, while the level of sICAM-2 was no significant change in ACIS group.2 The expression of CD42b was significant difference between patients with ACIS and controls individuals.3 The genetype frequencies of the VNTR gene polymorphism of GPIba in Kunming Han population are significantly different from Western population,partly different from other population in domestic regions.4 There was statistically significant difference in the frequency of CC and CD genotype between the patients with ACIS and the control group.5 The Logistic regression analysis revealed that elevated sICAM-1, sICAM-3, decreased CD42b and CC genotype of the VNTR gene polymorphism were the independent risk factor in ACIS.6 The expression of CD42b had no significant diferences between the all kind of VNTR gene polymorphism of GPIba.Conclusions 1 The expression of ICAM and CD42b and the CC genotype of the VNTR polymorphism in platelet membrane GPIba associated with ACIS.2 The genetype distribution of the VNTR gene polymorphism of GPIba in Kunming Han population are different from Western population and other population in domestic regions.3 The expression of CD42b are not affected by the VNTR polymorphism of GPIba gene.
