Objectives:To observe the expression of anti-cardiac myosin antibody and the myocardial remodeling in mice with chronic viral myocarditis (CVMG), and to compare the interventional effects of Captopril and Oxymatrine.Methods:The mice were randomly divided into six groups:normal control group1 (group 1) (n=8), normal control group2 (group 2) (n=8), CVB3 CVMC group1 (group 3) (n=20), CVB3 CVMC group2 (group 4) (n=25), captopril therapy group (group 5) (n=25) and oxymatrine therapy group (group 6) (n=25). Specifically, mice in group 3, group 4, group 5 and group 6 were inoculated with CVB3 at day 0,14 and 28 using 0.20ml, 0.25ml,0.30ml of CVB3, respectively. Mice in group 1 and group 2 received an equal-volume of normal saline without CVB3 at the 0,14th, and 28th day time points.Mice in group 1 and group 3 were measured by Echocardiography at day 42 after inoculation and weighted. Mice were anesthetized with 3% chloralhydrate intrapritoneally (0.01~0.015ml/g). Transthoracic echocardiography was using PHILIPS 7500 (Phillips) equipped with 3.5 MHz linear transducer, parasternal long-axis and short-axis views of the left ventricle were obtained to measure heart rate (HR), left ventricular end-diastolic internal diameter (LVDD), left ventricular end-systolic internal diameter (LVSD), peak velocity of aorta (Vp) at least 3 consecutive cardiac cycles. The ejection fraction (EF) and fractional shortening (FS) were calculated respectively. Mice were killed after echocardiographic analysis. Peripheral blood was obtained by eye-ball excision, and heart was resected for the following examination. Following the day 42, group 5 was treated with Captopril (100mg/kg) daily by gavage, group 6 was treated with oxymatrine (100mg/kg) daily by gavage for 28 days; concurrently group 2 and group 4 were received daily equal-volume gastric perfusion of normal saline. Mice were anesthetized and measured by Echocardiography at day 70. Mice were killed after echocardiographic analysis. Peripheral blood was obtained by eye-ball excision, and heart was resected for the following examination. The heart was weighed and the ratio of heart weight to body weight (HW/BW) was calculated. Heart slides were separated stained with haematoxylin-eosin (HE) and collagen specific Picric acid-Sirius red staining, and the collagen volume fraction(CVF) was calculated with image analysis software.
