AIM:The internal quality of the Chinese medicine was direct related to its efficacy and safety. It can not achieve the purpose of quality control of Chinese medicine by detecting single or a few components of the medicine generally. After the "fingerprint of TCM injections technical requirements (temporary)" was promulgated in 2000, the fingerprint had become to be a hot topic on quality of research and control of Chinese medicine. However, it was realized that the so-called fingerprinting confined to analysis of "chemical composition group", and the intrinsic quality and efficacy of Chinese medicine can not to be reflected accurately. The aim of this study was to build a relationship between "chemical composition group" and anti-tumor activity of traditional Chinese medicine.HPLC fingerprints of Zanthoxylum nitidum(Roxb.) DC was constructed base on two kinds of tumor cells 7901 and 7404 as efficacy model. And based on the anti-tumor activity and the establishment of HPLC fingerprint, "Chromatogram– effect "relationship of Zanthoxylum nitidum (Roxb.)DC. was constructed by backward technique.Method: (1) Anti-tumor active sites of Zanthoxylum nitidum (Roxb.)DC were selected and detectedFirstly, the reflux method was used to extracted different solvent extracts of Zanthoxylum nitidum(Roxb.) DC. by four kinds of solvent that included petroleum ether, chloroform, ethyl acetate and water, different solvent extracts were prepare. Secondly, the anti-tumor activity site was selected from the different solvent extracts by MTT method which through by two kinds of tumor cells 7901 and 7404 as efficacy model. Thirdly, all the samples's anti-tumor activity was deteced, and then inhibitory rate was calculated.(2) The HPLC fingerprint of Zanthoxylum nitidum (Roxb.)DC was establishedAccording to the requirements of chemical fingerprint, HPLC fingerprint of anti-tumor activity sites were established and evaluated by using chromatographic fingerprint evaluation system software, which was promulgated by the State Pharmacopoeia Commission.(3) The"Chromatogram– effect"relationship model of Zanthoxylum nitidum (Roxb.)DC. was establishedThe peak area in HPLC fingerprint was quantified and combined the anti-tumor activity of Zanthoxylum nitidum (Roxb.)DC. Anti-tumor "Chromatogram– effect" relationship of Zanthoxylum nitidum (Roxb.)DC. was established by using backward technique.(4) Contents of L-sesamin, nitidine chloride, 6-Ethoxychelerythrine in each sample was deteted by HPLC method. Combination the relationship of inhibitory effect on the tumor cell 7901 and tumor cell 7404 between these three chemicals concentration and then speculated that the three samples of the efficacy of chemical substances, try to describe interaction effect of each components in Zanthoxylum nitidum (Roxb.)DC.(5) The cluster analysis was used to identify The roots, stems of Zanthoxylum nitidum (Roxb.)DC. and Toddalia asiatica Lam. of the five origin.Results: (1) the anti-tumor active site of Zanthoxylum nitidum (Roxb.)DC was in the Chloroform extraction which contained alkaloids such like Nitidine chloride, 6-Ethoxychelerythrine. The anti-tumor activity of Samples were ranked highest to lowest were: Napo town>Rongshui town>Datang town>Wuming town>medicine market of Yulin.(2) In the HPLC-fingerprint of Zanthoxylum nitidum (Roxb.)DC. was stablished in this study. There were 24 fingerprint peaks of them included of known anti-tumor activity components such as Nitidine chloride, 6-Ethoxychelerythrine and so on. Established HPLC fingerprint of the distinction can be good in distinguishing the Zanthoxylum nitidum (Roxb.)DC from both sides non-medicinal parts and adulterants. This will help further standardize the quality standards for the Zanthoxylum nitidum (Roxb.)DC.(3) The results of backward technique showed that: 16 peaks in HPLC fingerprint had anti-tumor effect in 7404. The"Chromatogram– effect"relationship was that: Y=61.694+9.154X1-12.423X2-4.137X4+9.241X6-14.578X7-2.129X10-9.759X12 -4.819X13-11.475X15+5.356X16+4.041X18-6.399X19+4.209X20+16.245X22 -1.757X23+9.155X24(R2=0.923);there were 14 peaks in HPLC fingerprint had anti-tumor effect to 7901,"Chromatogram– effect"relationship was that: Y=79.153-8.503X1+6.79X5+4.529X6-5.108X7+5.231X10-8.32X11-9.404X12 -23.646X15+9.853X16+5.008X17-6.819X20+3.987X21+3.935X22-3.006X23(R2=0.913). It showed that the calculated value were very close to the actual value compared to the datas of 10 origins Zanthoxylum nitidum (Roxb.)DC.(4) The efficacy in single components of Zanthoxylum nitidum (Roxb.)DC. was less than the total efficacy of Zanthoxylum nitidum (Roxb.)DC; and the together efficacy of L-sesamin, Nitidine chloride, 6-Ethoxychelerythrine was greater than the total efficacy of Zanthoxylum nitidum (Roxb.)DC .(5) The Zanthoxylum nitidum (Roxb.)DC. roots, stems and Toddalia asiatica Lam. roots can be identifed by using cluster analysis methods.Conclusion: (1) Because Zanthoxylum nitidum (Roxb.)DC. have complex chemical compositions which directly affect its efficacy differences. The quality can not be controlled by a single or several ingredients of chemical composition.(2) The activity components such as L-sesamin, Nitidine chloride, 6-Ethoxychelerythrine of Zanthoxylum nitidum (Roxb.)DC. are better reflected through HPLC fingerprint. There also use to identify the authenticity of herbs which is reached the aim to control the quality and efficacy of herbs.(3) The"Chromatogram– effect"relationship can shows the quality of Zanthoxylum nitidum (Roxb.)DC. and guideline of clinical safety and effectiveness of clinical medicine.(4) Chemical composition in Zanthoxylum nitidum (Roxb.)DC. have promotion and inhibition effect, and the effects are the results of various components'unite.(5) The roots, stems of Zanthoxylum nitidum (Roxb.)DC. and Toddalia asiatica Lam. are different due to the chemical composition that can be easy to be identified by using cluster analysis method.
