Abstract Objective:Exploration part of the cytokine elevel of dendritic cell induced from peripheral blood monouclear cells,which taked suction from the patient of RRMS. Analysis the affect of DC and cytokine in immunopathogenesis of RRMS. Approach the influence of IFN-β1a in cytokine from DC.to reveal the mechanism treating RRMS from cytokine elevel.Method:Mononuclear cells were isolated from peripheral blood of RRMS patients (17 pou)and health control group (13 pou)by Ficoll-Hypaque density and cultured containing rhGM-CSF (100ng/ml),rhlI-4 (50ng/ml).Using LPS stimulates the mononuclear cells,which becamed mature,then adding IFN-β1a processes 48 hours. Eventually detect cytokine elevel of interleukin-12,23,27 and man tumor necrosis factor a.Results:The DC cultured in vitro turned into suspensive growth from adhesive situation on the 7th day,then the number of DC increased continuously and the cells showed the irregular morphologic appearance of DC.Flow cytometry showed that the percentages of CD86,CD80 on the blood-drived DCs from two sets were (90.4±5.2)% and (81.4±10.3)%. RRMS sources of DCs in the secretion of IFN-β1a before treatment, IL-12,23 and TNF-a increase, IL-27 in two groups of DCs in the treatment of secretion IFN-β1a no obvious difference, RRMS group after the DCs secrete IFN-β1a processing of IL-12, IL-23 and TNF-a significantly reduced, and the secretion of IL-27 reduce is not obvious, Control DCs secrete of IL-23 after treatment with IFN by former significantly reduced, and IL-12, IL-27 and TNF-a secretion decline is not obvious.Conclusion:1,Using rhGM-CSF and rhIL-4 induced people peripheral blood monouclear cells can get the high purity of mature DCs;2,RRMS, sources of DCs secrete more than normal control group of IL-12,23 and TNF-a, IL-12,23 and TNF-a in the pathogenesis of RRMS plays an important role, antagonism IL 12,23 and TNF-alikely as RRMS immune therapy targets; IL-27 in two groups of DCs in the treatment of secretion IFN-β1a no obvious difference, its role in RRMS is further research;3,After treatment, reduced IFN-β1a source of RRMS DCs secrete IL-12,23 and TNF-a, confirmed by IFN-β1a is cut IL-12,23 and the inflammatory factor of TNF-a in Th1/Th2 secretion, restore the dynamic balance in RRMS play therapy;...
