Objective:To explore the growth and proliferation of bone marrow hematopoietic stem/progenitor cell of myasthenia gravis(MG) in vitro. And to make a primary study of clinical application of hematopoietic stem / progenitor in myasthenia gravis.Methods: In vitro, both in 10 MG patients and 5 healthy controls,clones and proliferation of CFU-E and CFU-GM in bone marrow hematopoietic stem/progenitor cells were cultured with semisolid methycellulose, and CFU-MK with semisolid collagen .Results: (1) There were no significant differences in CFU-E,CFU-GM and CFU-MK colonies between MG patients and healthy controls respectively(p>0.05). (2) The colonies of CFU-E and CFU-GM in glucocorticoid of MG patients were less than healthy controls(p<0.05). The colonies of CFU-MK was less than healthy controls,but there was no statistical significance(p>0.05). Evermore,there was no significant difference in CFU-E,CFU-GM and CFU-MK between no-glucocorticoid of MG and healthy controls(p>0.05).(3) There was no significant difference in CFU-E,CFU-GM and CFU-MK colonies between glucocorticoid and no-glucocorticoid of MG(p>0.05).Conclution: (1)There was no obvious difference in CFU-E,CFU-GM and CFU-MK colonies of bone marrow hematopoietic stem/progenitor cell in myasthenia gravis. (2) The glucocorticoid could inhibit growth and proliferation of CFU-E,CFU- GM of bone marrow hemapoietic stem/progenitor cells in myathenia gravis ,but it has no significant effcet on CFU-MK. THE STUDY ON CELL CYCLE OF BONE MARROW MONONUCLEAR CELL IN MYASTHENIA GRAVISObjective:To investigate the changes of cell cycle-phase of bone marrow mononuclear cell in patients with myasthenia gravis preliminary, and to explore the role in pathogenesis of MG..Method:The cycle of bone marrow mononuclear cell in 5 myasthenia gravis patients were tested and analyzed by flow cytometry with PI staining method.Result:By contrast the cycle cell of BMMNC in myasthenia gravis patients and healthy controls,the proportion of phase G0/G1 and phase G2/M decreased, and the proportion of phase S and PI% increased.Conclusion:There maybe some abnormality in cycle cell of BMMNCs in patients.The BMMNCs of myasthenia gravis has more proliferative activity.
