Objective The aim of this study was to observe the level of IL-34 and IL-35 in peripheral blood mononuclear cells (PBMCs) with dilated cardiomyopathy (DCM),and investigate the relation of IL-34 and IL-35 level in PBMCs with DCM disease severity. Explore the role of IL-34 and IL-35 in human DCM.Methods Thirty cases of patients with DCM (DCM group),24 cases of heart failure patients (HF group) and 30 cases of normal adults (NC group) were studied. The subunit EBI3 (Epstein-Barr virus-induced gene 3) of IL-35 and IL-34 mRNA expression in PBMCs were detected by Reverse Transcription-Polymerase chain Reaction (RT-PCR). The protein levels of the two cytokines in plasma were measured with Enzyme-linked Immunosorbent Assay (ELISA).Results⑴IL-34 mRNA level or its protein level was significantly elevated in DCM patients compared with heart failure or normal control subjects.⑵IL-35 subuint-EBI3 or its protein level was significantly decreased in DCM patients compared with heart failure or normal control subjects.⑶The secretion of IL-34 was markedly correlation with the secretion of IL-35.⑷IL-34 protein level were strongly negatively correlated with ejection fraction (r =-0.450, P<0.01), and significantly positively correlatied with heart function ((r =0.589, P<0.01) of DCM patients.⑸The IL-35 protein level were strongly positively correlated with ejection fraction (r=0.449, P<0.05) of DCM patients, and were negatively correlated with heart function(r=-0.839, P<0.01) of DCM patients.Conclusion The ability to express IL-34 and IL-35 protein or mRNA in PBMCs is abnormal and the change strongly correlates with ejection fraction and heart function of DCM patients. It indicated that IL-34 and IL-35 may play an important role in the immune pathogenesis in DCM.
