Background & Objective Colorectal carcinoma is one of the most common malignancies with increasing incidence in the world. Its occurrence, pathogen- esis and progression are constantly changing. The changes in oncological mol- ecular biology have been confirmed as a new neo-index to distinguish metastasis and predict prognosis.ING1 is a recently cloned novel growth inhibitor and can- didate tumor suppressor gene . The main ING1 expression protein is p33ING1, which is related to the inhibition of cell growth , prolixferation ,cell aging, apop- tosis and negative regulation of checkpoint in cell cycle . The first discovered protein which stimulus tumor neovascularization is VEGF . In most cells , VEGF-C is recognized as the most important subtypes . Tumor neovasculariza -tion is an important element in tumor development and affects its biological behavior, in terms of invasion and metastasis. Metastatic cells can induce neo- vascularization feedback . So repeatedly, forming a progressive chain reaction. This paper was mainly researched on expressions and their significance of VEGF-C and p33ING1 protein in colorectal carcinoma.Methods Expression of p33ING1 and VEGF-C protein was investigated by Western-blot in 72 cases of colorectal carcinoma tissues ,tissues surrounding carcinoma and normal colorectal tissues (including 12 cases per Dukes,N1 and N2 stage). Using scanner to obtain the results of X-ray film .The average gray of objective bands and background in X-ray film were analyzed with Quanity One software. The former minus the latter as the statistical value. The final statistics for the sample values was each gray value with the corresponding ratio of gray values within the reference. Finally the data were analyzed by SPSS13.0 with the methods of ANOVA (analysis of variance),t test and Spearman.Results The results of this experiment confirm that relative expression levels of p33ING1 increase as following order: colorectal carcinoma tissues, surrounding carcinoma tissues and normal tissues (P<0.05). However, it is just opposite in the case of VEGF-C (P<0.05). The expression levels of p33ING1 and VEGF-C protein are related to Dukes stage and lymph node metastasis (P<0.05). There was negative correlation between the expression of p33ING1 and VEGF-C (P<0.05).Conclusion The expression of VEGF-C and p33ING1 were related to the onset and development of colorectal carcinoma. The detection of VEGF-C and p33ING1 with Western-blot may be of value in ca1culating the biological behaviors and the prognosis of colorectal carcinoma.
