ObjectiveIn this study,Two kinds of disinfectant, 0.5% iodophor and aiershi which containing effective chlorine 500mg / l were used during disinfection.we sample hospital wall oxygen supply device's oxygen connector as well as isogenesis humidified liquid before and after disinfection respectively. Bacterial cultures were done to the specimens sampled. To discuss the effect of disinfection from the positive rate of bacteria and changes of strain before and after disinfection. To show the importance of disinfection from the identity of bacterial species though in different parts. To provide a scientific basis if careful sterilization of the connectors is needed after the oxygen aspiration to prevent hospital infection.MethodChoosing 60 samples of unremovable part (Wall oxygen connector)of the wall oxygen supply device in the department of neurosurgery of our hospital. The samples were randomly divided into two groups, Each group of 30 cases,sample using the aseptic techniques。Disinfection method :The first group is sampled by 0.5% iodophor,disinfecting by using cotton swab containing iodophor,clean and polish the inner wall of oxygen connector thoroughly in spiral type。The second group is sampled by containing effective chlorine 500mg / l aiershi,containing effective chlorine 500mg / l aiershi disinfectant is used for disinfection which is filled in a bottle with piston vent,aim the spout vertically at oxygen connector lumen,sprayed two times from a distance of 2-3cm。Sample method : the method of oxygen connector before and after disinfection respectively。by sterile saline cotton swab which sample the inner surface of the oxygen connector in rotation, then shearing the part of cotton swab which contact the hand, and then placed the remaining part in the test tube which contains 10ml sterile saline, send it to be examined, make sure the whole process use aseptic technique. the Sample method of liquid wet, Diluted properly according to the degree of pollution, inoculated in ordinary nutrient agar plate, put them in the 37℃temperature box to culture 24h, select colonies needed to be identified using the blood and MacConkey agar plate which are incubated for 24 hours. To determine the relegation of bacteria initially by characteristics of bacterial colony growth and smear test under microscope after Gram stain of bacteria. Further, using Walkaway Identification System for identification of bacteria, For those whose coincidence rate is not very high ,API identification is needed。Making bacteriological identification from 60 cases of oxygen humidified liquid in the control group。Sample method: Drawing 1ml oxygen humidified liquid by sterile syringe and adding it to a tube containing 4ml nutrient broth, then send it to be examined immediately。Qualitative data, Adoptingχ~2 test for significance analysis of germ-carrying conditions in two groups. Using SPSS13.0 software package for data processing. P<0.05 is considered as significant difference.ResultsIn the department of neurological surgery and other sections,there are 17 cases of Petri dish covered with uncountable bacteria,13 cases are found to have more than three kinds of bacteria. Adoptingχ~2 test for significance analysis of germ-carrying conditions in two groups.χ~2=0.81,P>0.05,there is no statistically significant.The first group, After 0.5% iodophor disinfection in the experimental group, the bacteria-positive rate reduced remarkably by 73.33% and 16.67%, The second group ,After containing effective chlorine 500mg / l aiershi disinfectant in the experimental group, the bacteria-positive rate reduced remarkably by 76.67% and 26.67%, by theχ~2 test, P <0.01, it has statistical significance. Test the positive rate of bacteria after disinfection by two kinds of disinfectant by usingχ~2 test,χ~2 = 0.884, P>0.05, there is no statistically significant. 27 strains of bacterium are cultivanted from 30 samples in the first group is sampled by 0.5% iodophor, among the total, there are G~+ coccus,G~- coccus,G~+ bacilus,G~- bacilus , G~+ bacilus account for 59.26%,G~- bacilus account for 22.22%.,G~+ coccus account for 11.11%,G~- coccus account for 7.41%。32 strains of bacterium are cultivanted from 30 samples in the first group is sampled by 500mg / l aiershi, among the total, there are G~+ coccus,G~- coccus,G~+ bacilus,G~- bacilus ,G~+ bacilus account for 65.63%,G~- bacilus account for 21.88%.,G~+ coccus account for 9.38%,G~- coccus account for 3.13%。From the constituent ratio, G~+and G~- bacilus are main constitution. Two kinds of bacteria species change after the disinfectant,After sterilization by 0.5% Iodophors of the first group, species decrease from 9 to 2, strains decrease from 27 to 5, remaining strains are Bacillus subtilis and Pseudomonas alcaligenes. Checked byχ~2 test, the difference is nonsignificant. After sterilization by airshi of the second group, species decrease from 9 to 2, strains decrease from 32 to 8, remaining strains are Bacillus subtilis and staphylococci Epidermidis. Checked byχ~2 test, the difference is nonsignificant. Identification results of liquid oxygen species of bacteria in the wet :17 strains of bacterium are cultivanted from 30 samples in the first group is sampled, among the total, there are G~+ coccus,G~- coccus,G~+ bacilus,G~- bacilus , G~+ bacilus account for 29.41%,G~- bacilus account for 41.18%.,G~+ coccus account for23.53%,G~- coccus account for 5.88%。19 strains of bacterium are cultivanted from 30 samples in the second group is sampled, among the total, there are G~+ coccus,G~- coccus,G~+ bacilus,G~- bacilus , G~+ bacilus account for 42.11%,G~- bacilus account for 31.58%.,G~+ coccus account for 21.05%,G~- coccus account for 5.26%。By the classification of gram's staining, compare the strains of bacterium of oxygen connector as well as oxygen humidified liquid,Checked byχ~2 test, P>0.05,the difference is nonsignificant. 27 strains of bacterium are cultivanted at the connectors of the first group, 9 strains of them are same as oxygen humidity liquid, strains changes in the two sites are checked byχ~2 test,P>0.05, difference is nonsignificant.the identical strains are Staphylococcus epidermidis, enterococcus, Neisser's coccus, Bacillus subtilis,E coli, Pseudomonas aeruginosa。strains changes in the second group are checked byχ~2 test,P>0.05,there is statistical significance.Staphylococcus epidermidis, saprophytic staphylococci, enterococcus, Bacillus subtilis,Bacillus cereus, Proteus, Pseudomonas stutzeri, Escherichia coli。Conclusion1. Disinfection can make the bacteria positive rate of wall oxygen supply device connector mouth devices lower.2. Identity of bacterial species exist in the wall oxygen supply device connector mouth and oxygen humidified liquid.3. Disinfection can reduce the the bacterial quantity of wall oxygen supply device to connect the mouth effectivly, there is no significant difference compared with the ones before disinfection of bacterial species, there is a result of study that disinfection the inner wall of oxygen connector 0.5% iodophor better than containing effective chlorine in clinical practical work.
