Objective: To study the effects of different concentrations of Pseudolaric acid B on human retinal vascular endothelial cells(HRCECs)proliferation and vascular endothelial growth factor(VEGF)expressing in high glucose environmentMethods: 1.Human retinal microvascular endothelial cells(HRCECs)were cultured in vitro,and cells in logarithmic phase were selected for the experiment.The experimental groups were divided into low glucose group,high glucose control group,and high glucose + 2umol/L of Pseudolaric acid B group concentration(P1),high glucose+ 4umol/L of Pseudolaric acid B group concentration(P2),high glucose+ 8umol/L of Pseudolaric acid B group concentration(P3).2.The inhibition of cell proliferation were detected in different concentrations of Pseudolaric acid B in high glucose environment with HRCECs MTT method.3.With cell Western Blot method,HRCECs the expression of VEGF were detected when the different concentrations of Pseudolaric acid B in vitro were being cultured under high glucose environment.Results:1.While HRCECs cells in high glucose environment were being cultured for 12,24,36 hours,MTT test results showed that the absorbance value of cells in PH group,compared with PL group,increased at the same time.The difference was statistically significant(P < 0.05).As time goes on,the absorbance value of cells in PH group increased.Compared with PH,the absorbance values in group P1,P2,P3 were decreased,the difference was statistically significant(P < 0.05).And with the increase of drug concentration and the decrease of the absorbance value,the absorption of the same concentration of Pseudolaric acid B group were compared with each other at different time points.The difference was statistically significant(P < 0.05).As time goes on,the absorbance value decreased.At the same time point,the absorption of the difference concentration of Pseudolaric acid B group was compared with each other.The difference was statistically significant(P < 0.05).And with the increase of the concentration,the value of absorbance decreased.2.While HRCECs cells in high glucose environment were being cultured for 12,24,36 hours,they were detected.Western Blot method results showed that there is a few expressions of VEGF in cells in PL group at three different time points.The expression hadn't significantly increased,so the difference was not statistically significant(P < 0.05).Compared with PL group,the VEGF level of the cells in the PH group was increased,and the difference was statistically significant.As time goes on,VEGF gray value in PH group increased.Compared with group PH,the grey value of VEGF in the P1,P2,P3 group were decreased.The difference was statistically significant(P < 0.05).With the increase of drug concentration,the gray value of VEGF was lowering.At different time points,the gray value of VEGF in the same concentration of Pseudolaric acid B group compared with each other,and the difference was statistically significant(P < 0.05).As time goes on,the gray value decreased.At the same time points,the gray value of VEGF in the different concentration of Pseudolaric acid B group were compared with each other,and the difference was statistically significant(P < 0.05).And with the increase of concentration,the gray value became lower.Conclusion:1.Pseudolaric acid B can inhibit the proliferation of HRCECs in vitro high glucose environment,and was of time and concentration dependent.2.Pseudolaric acid B can be reduced under high glucose condition in vitro growth of HRCECs expression and the content of VEGF,and also is of time and concentration dependent.3.Pseudolaric acid B can inhibit the proliferation of HRCECs in high glucose environment may be achieved by lowering the expression of VEGF in HRCECs.
