ObjectiveTo investigate the preventive effects of Quchiling on mise with Alzheimer's disease (AD) induced by administration of D-galactose (D-gal) and of Aluminum trichloride (AICI3).Method(1) The mice were randomly divided into six groups:blank group, model group, Quchiling low, moderate and high dose groups, and the positive drug control group of Huperzine A;Except the blank group, The mice were treated with intraperitoneal administration of D-gal ]20mg·kg-1·d-1and intragastric administration of AICI3 20mg·kg-1·d-1 for 10 weeks; the therapy groups oral administration Quchiling with concentration of (2.3g·kg-1·d-1,4.6g-kg-1·d-1, 9.2g·kg-1·d-1) from the 7th week to 10th week. (2) The abilities of learning and memory were assessed by Morris water maze test and step down test. The expression of amyloid beta-protein precursor (APP) and the content of tau protein were analyzed by reverse transcription-polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA) respectively. Hematoxylin-eosin (HE) staining, neurofibrillary tangles (NFT) staining and amyloid beta protein (Aβ) immunohisto chemistry were applied to detect the pathological changes in brain.Results(1) ethology test:compared with model group, the escape latency of Quching-treated groups was shortened, the wrong times of step down test were significantly decreased(P<0.05), the number of times that mice crossed the original platform location and the percentage of swimming time spent in the target quadrant were significantly increased (P<0.05), the high dose of Quching gave a significant performances (P<0.01);(2) biochemical indicator:compared with model group, The expression of APP mRNA in Quching-treated groups were significantly decreased(P<0.01). However, there were no significant differences about tau protein among all groups. (3) compared with model group, the neurons in cortex and hippocampus were intact and arranged regularly and the dyeing was uniform and stable in neuron staining in Quchiling groups. The number of neurofibrillary tangles and senile plaque was decreased in the group of Quchiling.Conclusion(1) The mise model of AD induced by D-galactose and Aluminum trichloride was credible.(2) Quchiling could prevent the AD.
