Liriope Lour. are perennial herbaceous plants in the Liliaceae family, mainly distributing in southern China. The tubers of Liriope Lour. are used in Chinese folk medicine. Liriope is recorded in China Pharmacopoeia (1995 edition) including Liriope spicala (Thunb.) Lour. var.prolifera Y.T.Ma and Liriope musacarli (Decne.) Bailey. As a tonic to moisten yin, it has been employed in traditional Chinese medicine for treatment asthma, bronchial and lung inflammation. Due to more and more bioactive substances isolated from this family, such as steroidal saponins and polysaccharides, the plants of Liriope Lour. have become the subject of continueing and growing interest. No any phytochemical investigation has been done on the species of Liriope graminifolia (L.) Baker. On our searching for plant-derived bioactivities compounds, the plants of Liriope graminifolia (L.) Baker were chemically studied.8 compounds were isolated, purified from this species. Their structures were determined by UV, MS, ID and 2D NMR analysis, respectively, all of them are first isolated from this plants, and two of them are new compounds. It provided the experimental data for the further development of Liriope graminifolia (L.) Baker.1. The advances in isolation and bioactivities of components from Liriope Lour. were reviewed, which can provide the basement for the research of Liriope graminifolia (L.) Baker.2.The chemical constituent in the ethyl acetate extract of Liriope graminifolia (L.) Baker. was separated by chromatography on silica gel, Sephadex LH-20 and Pre-HPLC to yield three compounds: methylophiopogonanone B (compound 1),5,7-dihydroxy-6-methyl-3-(4-methoxybenzyl)-chroman-4-one (compound 2) and 7,4'-dihydroxy-5-methoxy flavanones (compound 3).3.The chemical constituent in the n-butanol extract of Liriope graminifolia (L.) Baker. was isolated by D101 macroporous adsorption resin eluted with water,25%,50%,75%and 95%ethanol to obtain the components with different polarity. The fraction of 50%ethanol was isolated by medium pressure liquid chromatograph (MPLC), Sephadex LH-20 to obtain hesperidin (compound 4) and 25(R, S)-ruscogenin-l-O-sulfate-3-O-a-L-rhamnopyranoside (compound 8). The fraction of 75% ethanol was isolated by silica gel to obtain three compounds: 25(S)-ruscogenin-l-O-β-D-xylopyranoside-3-O-a-L-rhamnopyranoside (compound 5), 25(R)-ruscogenin-1-O-α-L-rhamnopyranosyl(1→2)-β-D-xylopyranoside (compound 6), (25S)-spirost-5-ene-3β,17α-diol-3-O-β-D-xylopyranosyl( 1→3)-α-L-arabinofuranosyl(l→2)-[α-L-rhamnogyranosyl (1→4)]-β-D-glucopyranoside (compound 7). Compound 6 and 7 were new compounds.4.Anti-tumor biological activity of compounds from Liriope graminifolia (L.) Baker: By anti-tumor biological activity in vitro screening test (MTT method), compound 1 showed strong anti-tumor activities against human cervical carcinoma (Hela, IC50=6.0±2.4μg/ml); new compound 7 had moderate anti-tumor activities against human cervical carcinoma (Hela, IC50=26.1±4.4μg/ml) and human hepatocellular carcinoma (SMMC-7721, IC50=76.4±20.6μg/ml); compound 5 had antiproliferative activity to human cervical carcinoma (Hela, IC50=13.3±3.0μg/ml)。The results of present study will be useful to further phytochemical investigation on Liriope graminifolia (L.) Baker.
