Ovarian carcinoma is one of the most common gynecological tumors. The tumor is difficult to perceive at early stage, because it has an insidious onset. It is usually detected in late clinical stage, when a majority of tumors have metastasis. At that time, it is difficult to eradicate the tumor by surgery, since the tumors are frequently transferred to other areas, and the tumors will relapse after the operation. Therefore, ovarian carcinoma is one of gynecological malignancies with poor prognosis. It is urgent to further study the transfer and relapse mechanism of primary ovarian carcinoma and to detect the effective treatment measures in the diagnosis and treatment of primary ovarian carcinoma.E-cadherin, which makes the main function during adhesion between the polarized epithelia, is one of the main members of cadherin family. Expression of E-cadherin is specific in different tissues and organs. As a metastatic suppressor molecule in carcinomas, the decrease expression of E-cadherin is not only associated with tumour progression and metastases formation in a series of different cancers, but also results in the development of cancer by activating the WNT/β-catenin signal transduction pathway and specifically promoting over-expression of oncogenes such as c-myc, cyclinD1 and so on. E-cadherin may be an important target in the treatment of metastatic ovarian.Claudins is a kind of skeleton proteins that can make the tight junction of maintaining the specific barrier function. Expression of Claudins is highly tissue specificity. Tumorigenesis usually accompanies the damage of tight junction, which makes enhancement of invasiveness and attenuation of intercellular adhesion. Research showed that expression of Claudins is probably related to differentiation grade and invasiveness in various epithelial malignant tumors. Expression of Claudin-3 could promote motility, invasiveness and survivability.In this experiment, we take a study of the expression level of Protein and mRNA of E-cadherin and Claudin-3 in human epithelial ovarian tumor, and the relationship between In this study, we will use IHC and RT-PCR to detect the protein and mRNA level of E-cadherin和Claudin-3 in ovarian cancer. Futhermore, we will detect whether the expression levels of E-cadherin和Claudin-3 in ovarian cancer have some relationship with the results and clinical pathological parameters such as age, pathological type, TNM staging, pathological grade, peritoneal metastasis and so on. And we try to find whether CA125 is consistent with diagnosis of ovarian tumor by understanding the relationship between expressions of E-cadherin and Claudin-3 and invasion in addition to metastasis. Finally we try to provide evidence for clinical diagnosis and prognostic evaluation.Methods:1,To detect the expressions of E-cadherin and Claudin-3 using S-P immunohistochemistry in histological section of ovarian tumor.make statistical analysis of clinical information of 64 patients.2,By using Real-time PCR to detect the gene expressions of E-cadherin and Claudin-3 in normal ovary tissue and benign tumor and ovarian cancer.3,Make statistical analysis of clinical information of patients.Results:1,Expression of E-cadherin and Claudin-3 in normal and epithelial ovarian tumor:(1) The positive staining of E-cadherin are 35 cases, with positive rate 54.7%(35/64). E-cadherin do not be expressed in normal ovarian tissues, while expressions of E-cadherin in ovarian tumor were significantly lower in malignant group than those in benign group and borderline group(P<0.01).(2) The positive staining of claudin-3 are 40 cases, with positive rate 62.5%(40/64) Claudin-3 do not be expressed in normal ovarian tissues, while expressions of Claudin-3 in ovarian tumor were significantly higher in in malignant group than those in benign group and borderline group (P<0.05).2,Gene expression of E-cadherin and Claudin-3 in normal ovary tissue and benign tumor and ovarian cancer.(1) E-cadherin mRNA gene expression in ovary benign tumor and the stageⅠ,Ⅱ,Ⅲand IVof ovarian cancer are the 0.950.92,0.736,0.534 and 0.346 times to the normal one. (P<0.05)(2) claudin-3 mRNA gene expression in ovary benign tumor and the stageⅠ,Ⅱ,ⅢandⅣof ovarian cancer are the1.5,2.12,2.99,14.56 and 19.22 times to the normal one. (P<0.05)3,E-cadherin and claudin-3'expression with epithelial ovarian tumor clinic clinicopathological features(1) A significant correlation is found between expressions of E-cadherin and FIGO staging, pathological type, pathological grade, survival of patients and peritoneal metastasis(P<0.01). However it is unrelated to age and CA125 (P>0.05).(2) A significant correlation is found between expressions of claudin-3 and FIGO staging, pathological type, pathological grade, survival of patients and peritoneal metastasis(P<0.01). However it is unrelated to age and CA125 (P>0.05).4,E-cadherin and claudin-3'expression with the prognosis of patientsStatistical analysis shows that the expressions of E-cadherin and claudin-3 has significant correlation with the prognosis of patients (P>0.05).Conclusion:1,Down regulation of E-cadherin with up-regulation of Claudin-3 in epithelium tumor of the ovary to emphasize significant effect.2,The expressions of E-cadherin and Claudin-3 correlate with the development, invasion and metastasis of epithelial ovarian tumor. Simultaneous detection expression may be useful for the prediction of diagnosis treatment in epithelial ovarian tumor.3,The E-cadherin and claudin-3 together with cell adheren,they get negative correlation in ovarian cancer, They may get synergistic action with each other.
