Objective On the basis of the assumption of the double receptor, the gene chip was taken advantage of in the experiment. Genes expression specific to OPIDN in the nerve of hens exposed to TOCP were screen.Methods 42 roman hens were divided two parts. The first part include 24 hens, randomly divided to 4 groups, control,1000 mg/kg TOCP,750 mg/kg TOCP and 40 mg/kg PMSF+1000 mg/kg TOCP. Six hens were divided in one group. Hens were sacrificed after 21 days. The second part include 18 hens, randomly divided to 3 groups, control,1000 mg/kg TOCP and 40 mg/kg PMSF+1000 mg/kg TOCP. Six hens were divided in one group. Hens were sacrificed after 7 days. Their cerebrum and lumbar were dissected and homogenized. Total RNA in the cerebrum and the lumbar was extracted, purifled, and then transcribed to cRNA probe. Then cRNA probe was biotin labeled and hybridized with Chiken Genome 430 2.0 Araay. Hybridization signals were scaned and gene chip data was analyzed.Results All hens in the 1000 mg/kg TOCP group were abserved OPIDN. 40 mg/kg PMSF can restrain the OPIDN of the hens exposed to 1000 mg/kg TOCP.37894 and 17026 genes were shown in the cerebrum and the lumbar. On the basis of the assumption of the double receptor,115 and 100 genes specific to OPIDN were abserved in the cerebrum and the lumbar. According to the molecule function, the genes specific to OPIDN were divided 8 sorts, including to protein binding, transporter activity,protein phosphorylation, ion channel activity, catalytic action, transcription and regulation, nucleotide binding and others.In the cerebrum, SLC9A2.SLC47A2,SLC25A14,SLC19A1,GRIN2B, PDE6B,PDE5A,PDE1A and PDE10A genes was regulated up 2.14,2.27,3.18,2.64,2.74,26.75.2.32,2.31 and 2.38 times; SLC16A10 and TUBB1 genes was regulated down 3.33 and 3.32 times. In the lumbar, SLC9A2 and SLC25A14 genes was regulated up 2.50 and 3.67 times; SLC16A1,SLC35A3 and SLC26A8 genes was regulated down 2.47,2.74 and 4.93 times.Conclusion 115 and 100 genes specific to OPIDN were abserved in the cerebrum and the lumbar. The genes of SLCs and PDEs were relative to OPIDN.
