Background and aims:Ulcerative colitis(UC) is a chronic inflamma-tory bowel disease with unknown etiology.It was a common disease in Western countries, but the incidence rate was gradually increased in China in recent years. Present researches showed that hereditary factors, environmental factors(infection) and immunity contributed to the occurrence of UC.The altered immunity in the intestines played an important role in the progress of the disease.Denfensin is one of the antibacterial peptides in the body and is the component of the intestinal innate immunity.The aim of this study was to detect the expression of HNP1-3 in patients with UC, to investigate the significance of HNP1-3 in the progess of UC.Methods:15 UC patients were entrolled in the study who were from the First Affiliated Hospital of Dalian Medical University and the Xinhua Affiliated Hospital of Dalian University during Oct.2009 to Feb.2010. Among these patients,9 were males and 6 were females with the age ranged from 20 to 81 years old,all of them were in the active period.The samples were taken from the inflamed area and non-inflamed area(10cm away from the inflamed mucosa) during colonscopy exam.According to the colonscopy exam,4 patients were classified into gradeⅠ,7 into gradeⅡ,4 into gradeⅢ;according to the clinical manifestation,5 patients were classified into mild type,7 into moderate type and 3 into severe type;according to the pathology,3 patients were classified into grade 0-Ⅰ,9 into gradeⅡ,3 into gradeⅢ-Ⅳ. Based on the inflamed location,there were 3 patients with rectum lesions,6 with rectosigmoid lesions,6 with hemi-colon lesion.The immunohisto-chemistry method was used to detect the HNP1-3 in the tissues.The positive expression of HNP1-3 was compared between the inflamed group and the noninflamed group,also was compared among different grades and different inflamed locations in the the inflamed group.The deta were analyzed with SPSS 17.0 software.Results:What we see under the microscopy was that there were lots of inflammatory cells infiltrating in the tissues of the patients with active UC,HNP1-3 was mainly expressed in neutrophils with a little expression in the epithlial cells. The positive rate of the inflamed group was 75%and the rate of the non-inflamed group was 40%.The expression of HNP1-3 in the UC inflamed group was significantly higher than that in the non-inflamed group(P<0.05).In the UC inflamed group,the degree of HNP1-3 positive expression was significantly different in different grades according to the colonscopy examination(P<0.05),the degree of HNP1-3 expression was significantly different in different grades according to the pathology (P<0.05), the degree of the HNP1-3 expression showed no significant difference in different groups according to the clinical manifestation (P>0.05),the degree of the HNP1-3 expression showed no significant difference in different inflamed lesions(P>0.05).Conclusion:1.The expression of HNP1-3 in the UC inflamed area is greatly increased, so that HNP1-3 may participate in the development of UC.2.The degree of HNP1-3 expression isn't correlate with the clinical manifestation,but the degree of HNP1-3 expression is greatly parallelly increased with the increasing grades both in pathology and coloncopy, this indicates that the degree of HNP1-3 expression can reflect the severity of UC in some extent.3.The expression of HNP1-3 isn't correlate with the inflamed locations, so that HNP1-3 can't reflect the scopes of UC.
