Objectiveâ‘ Research the second window of protection of sevoflurane preconditioning against myocardial ischemia/reperfusion injury in rats in vivo.â‘¡Investigate the role of autophagy in the mechanism of SWOP of sevoflurane preconditioning.Methodsâ‘ Through the determination of infarct size and examination the tissue injury of myocardium with optical microscope, research the second window of protection (SWOP) of sevoflurane preconditioning against myocardial ischemia/reperfusion injury in rats.â‘¡Measure LC3-â…¡by Western Blot analysis before myocardial ischemia and after myocardial ischemia reperfusion, and observe the morphology of autophagy with transmission electron microscope.Resultsâ‘ Infarct sizes were reduced in SWOP group compared with I/R group.â‘¡Light-Imprisoning Mirror: The samples obtained after reperfusion were all injured, but SWOP group was less injury than I/R group.â‘¢Electric Mirror: The number of autophagosomes before ischemia in SWOP group were more than I/R group, after reperfusion there were observed a lot of autophagosomes and lysosomes in I/R group and SWOP group, but there were a few in SWOP group than that of I/R group .â‘£The levels of LC3-â…¡and Cathepsin B were all significantly up-regulated in SWOP and SEVO group before ischemia in rat myocardium as compared with the CON group. The levels of LC3-â…¡, and Cathepsin B were all significantly up-regulated in I/R and SWOP group at the end of reperfusion in rat myocardium as compared with the CON group, but the levels of LC3-â…¡was much lower in SWOP group than in I/R group.Conclusion Sevoflurane preconditioning maybe inhibit the excessive activation of autophagy at the end of reperfusion through properly up-regulating autophagy expression before ischemia, reduce myocardial infarct size in rats, decrease the myocardial tissue injury, and then produce he second window of protection.
