Objective:To evaluate the effect of GSK-3? in the diabetic factor attenuates the myocardial protection of sevoflurane in rats.Method:Healthy clean male SD rats,weighing 210-300 g,establishing diabetic rat mod el by single intraperitoneal injection of streptozotocin(STZ)60 mg/kg,choose 12 r ats in which to be the group of DS(diabetic rats with ischemia-reperfusion and se voflurane post-procession).Another 48 healthy SD rats that received simultaneous fe eding are divided into four groups(n=12)according to the principle of random contr ol:group Sham(sham operation),group NI(ischemia-reperfusion),group NS(ischem ia-reperfusion and sevoflurane post-procession),and group GS(ischemia-reperfusion and GSK-3? overexpression).Ischemia-reperfusion model was induced by occlusion of the left coronary artery for 30 min followed by 120 min reperfusion.The post-p rocession of sevoflurane was followed by inhalation of 2.5% sevoflurane for 5 min from 1 min before reperfusion.GSK-3? overexpression was performed by tail-vein injection of GSK overexpressing recombinant adenovirus(AAV9)one week before t he ischemia-reperfusion model was constructed.The ECG performance of myocardia l ischemia was observed in the preparation of ischemia-reperfusion model in each g roup(stern elevation,large Q wave and arrhythmia).Abdominal aortic blood was e xtracted and the concentration of cTnI was tested after 120 min.And then executed the rats,took the heart,measured myocardial infarct size and myocardial pathologi cal damage.Detecting the expression of GSK-3? and caspase-3 by Western-blot.Result:Compared with Sham group,the concentration of cTnI and the infarct size wer e significantly increased,and the expression of GSK-3? and caspase-3 were up-regu lated in the group of NI,NS,DS and GS(P <0.05).Compared with NI group,the concentration of cTnI and the infarct size were decreased,and the expression of GSK-3? and caspase-3 were dowm-regulated in the group of NS(P <0.05).Compar ed with NS group,the concentration of cTn I and the infarct size were significantly increased,and the expression of GSK-3? and caspase-3 were up-regulated in the g roup of DS and GS(P <0.05).Compared with DS group,no significant change wa s found in the parameters mentioned above in group GS.(P >0.05).Compared with Sham group,the sigh of myocardial ischemia was appeared on the ECG and the pathological damage was increased in the group of NI,NS,DS and GS.Compared with NI group,the ECG performance of myocardial ischemia w as not obvious and the pathological damage was decreased in the NS group.Comp ared with NS group,the ECG performance of myocardial ischemia appearing and t he pathological damage increasing in the DS and GS group.Compared with DS gr oup,the difference of ECG performance and pathological damage are not obvious.Conclusion:Overexpression of GSK-3? can aggravate myocardial I/R injury in rats,and the decreased protection of sevoflurane postconditioning on myocardial I/R in diabetic rats may be caused by GSK pathway...
