Objective①Research the SWOP of sevoflurane preconditioning against myocardium ischemia/reperfusion injury.②Research the effect of NF-кB during SWOP, then evaluate the status of p65 and p50 in this mechanism.Methods①Set up the model of myocardial ischemic-reperfusion in vivo, observe the effect of SWOP through determing myocardial infarct size.②Measure the expression of NF-кB (p65, p50) and p-IκBαbefore ischemia and after reperfusion through interfering with PTN, NF-κB inhibitor.Results①Sevoflurane significantly reduced infarct size, and this effect could be blocked by PTN.②Sevoflurane up-regulated the expression of NF-кB (p65, p50) and p-IκBαproteins compared to that of the Control group before ischemia.③The expression of NF-кB (p65, p50) and p-IκBαproteins were up-regulated in Simple-Ischemic and SWOP group compared to that of Control group after reperfusion, but the up-regulated adjustment of SWOP group was smaller than that of Simple-Ischemic group.Conclusion①Preconditioning with sevoflurane reduces myocardial infarct size, produces SWOP.②It is verified that NF-κB participates in the mechanism of SWOP through interfering with PTN, NF-κB inhibitor.③NF-κB through negative feedback control mechanism may play an important role in the mechanism of SWOP of sevoflurane preconditioning on myocardium against ischemia-reperfusion injury, p65 and p50 play the similar role in this mechanism.
