Objective:To determine the Alteration ofα1-AR and Gq/11-protein in smooth musclein rats after partial unilateral ureteral obstruction.Method:Eighty Wistar rats had been divided into four groups by random:8-week experimental group (n=20) and sham control group (n=20),16-week experimental group (n=20) and sham control group(n=20). Experimental animals'left ureters (n=40) had been embedded the upper half into the psoas muscles. Sham control rats only received laparotomy. After the model was established the rats were separated in different experimental time point to examine theα1-AR of each group by direct immunofluorescence method,analyse the average gray value of fluorescence photographs in each group, compare theα1-AR expression between the different groups. At the same time using the western blot to examine the change of Gq/11-protein and compare the Gq/11-protein expression between the different groups-Results:There was a significant increase at the average gray value ofα1-AR in the 8-week experimental group compared with sham control group(P< 0.05). There was a significant decrease at the average gray value ofα1-AR in the 16-week experimental group compared with sham control group(P<0.05). There was a significant increase at the average gray value ofα1-AR in the 8-week experimental group compared with the 16-week experimental group (P<0.05).There was no significant difference at the average gray value ofα1-AR between the 8-week control group and 16 weeks control group (P>0.05). There was a significant increase at the average gray value of Gq/11-protein in the 8-week experimental group compared with sham control group(P< 0.05). There was a significant decrease at the average gray value of Gq/11-protein in the 16-week experimental group compared with sham control group(P<0.05). There was a significant increase at the average gray value of Gq/11-protein in the 8-week experimental group compared with the 16-week experimental group (P<0.05).There was no significant difference at the average gray value of Gq/11-protein between the 8-week control group and 16 weeks control group (P>0.05) Conclusion:It Demonstrates an increase ofα1-AR and Gq/11-protein in smooth muscl in PUUO rats at eight weeks while an decrease at sixteen weeks and it provides a molecular mechanism for dynamic changes of the increased smooth muscle contractility at early stage,while the decreased at late stage in PUUO.
