The source of Radix Codonopsis was various, which were widely located geographically. The germplasms of original plant was complex, and the morphous of the plant and the character of the medical material were aberrance, which resulted in poor medicinal quality. Due to the lack of the unified evaluation system of the medicinial materials, and the uneven quality of the medicines, effective control of quality of Radix Codonopsis was a significant problem that we have to resolve. As the chemical composition of Radix Codonopsis was complex and the element of main ingredients was low, the establishment of a more scientific chemical quality controlling model becomes a difficult issue. In this paper, the parallel comparison method was used, unified cultivation and management were carried out on Radix Codonopsis of different habitats. Based on research of the medical materials collected from Shanxi province of Lingchuan Radix Codonopsis GAP planting base of herbs and different habitats, the change regularity of chemical contents of Radix Codonopsis, as well as its relationship with the environment were disussed through the study of contents determination and fingerprint. The quality of Radix Codonopsis from different origins was compared, providing scientific basis for further standardized production of Radix Codonopsis. This paper used melting method to prepare polysaccharide from Radix Codonopsis drop pill. Studied on the influencing factors then found the best technics with orthogonal test. The quality standard of the drop pills was researched in this paper and the determination of polysaccharide from Radix Codonopsis was established.The main contents of this paper were summarized as follows:1. Assay of lobetyolin and atractylenoide III in Radix Codonopsis of different habitat.Assay Radix Codonopsis of different habitats and Radix Codonopsis of different habitats being introducted to ShanXi GAP planting base by the HPLC method, whith lobetyolin and atractylenoide III as indicators.To screen the two chemical constituents of lobetyolin and atractylenoide III in Radix Codonopsis of different habitats and Radix Codonopsis of different habitats being introducted to ShanXi GAP planting base for investigating environmental factors on the chemical composition of Radix Codonopsis. For atractylenolideⅢthe linear rang was 0.10-1.6ug (r=0.9994).The average recovery was 103.0%with RSD of 0.02%(n=6).For lobetyolin the linear rang was 0.01-2ug (r=0.9999).The average recovery was 100%with RSD of 2.1%(n=6).The result showed that there were difference in the contents of lobetyolin and atractylenoide III in Radix Codonopsis from different habiats. After being introduced, the contents of atractylenolide III all increased, the contents of lobetyolin in Radix Codonopsis from BianQiao and LongXi increased. Environmental conditions had a significant influence on the chemical composition of Radix Codonopsis. The natural environmental conditions of Shanxi were suitable for the cultivation and introduction.2. Assay of polysaccharide from Radix Codonopsis and extract in Radix Codonopsis from different habitatThis paper assay of polysaccharide used the method of phenol-sulfuric and the calculation of polysaccharide by the content of glucose. Calculated the ethanol extract using the method of hot-dip from pharmacopoeia. The content of polysaccharide from in was Radix Codonopsis from genuine habit of ShanXi and HuBei exceed 60%, from LongXi was 47%. The content of ethanol extract was all higher than the Pharmacopoeia requirement.This paper used the method of hot dipping from pharmacopoeia to get the extract content. The results also showed that Radix Codonopsis from genuine habit of ShanXi and HuBei were the best, and LongXi was fllowed.3. HPLC fingerprint of Radix Codonopsis from different habitatHPLC fingerprint of Radix Codonopsis was established, by the HPLC method, with 220nm being the wavelength of the detection of fingerprints, acetonitrile-water (0.1% phosphoric acid) as a linear gradient elution of the mobile phase. Lingchuan Radix Codonopsis from Shanxi Lu lingchuan GAP base being used as a reference map, glycoside peak (S peak,) as the control peak,11 peaks were flexed, and peak 4 and 5 were respectivelyβ-D-Fructose-butanol glycosides and syringin (see Figure 3-1). A comparative analysis was carried out on 10 different habit Radix Codonopsis cultivated on the the same habit in Shanxi lingchuan Codonopsis GAP base and 7 different original habit Radix Codonopsis. The results showed that glycosides,β-D-fructose-butanol glycosides and syringin were all important characteristic ingredients in Radix Codonopsis. HPLC fingerprint that contained 11 fingerprint peaks can be used as identification standards for the qualitaty of Radix Codonopsis, lobetyolin being used as the reference. Through the HPLC fingerprint clustering analysis of Radix Codonopsis, Radix Codonopsis from different habitats can be identified to a certain extent.4. Study on chemical constituents of Radix Codonopsis:The chemical constituents of Codonopsis had been studied in this paper through a variety of separation techniques (silica column chromatography, Sephadex LH-20 gel column chromatography and HPLC), combined with modern techniques (UV, ESI-MS, NMR). Four compounds were identified from Radix Codonopsis. They were stigmasterol, lobetyolin, syringin andβ-D-fructose-butanol glycoside. Among them, it was identified that lobetyolin, syringin and P-D-fructose-butanol glycoside were characteristic elements in Radix Codonopsis.5. Study on polysaccharide from Radix CodonopsisThis paper used melting method to prepare polysaccharide from Radix Codonopsis drop pill, study on the influencing factors:the velocity of dropping, the temperature of melting, diameter of burette, the ratio of medicament and PEG4000,PEG6000, then find the best technics with orthogonal test:the ratio of drug and PEG4000,PEG6000 was 1:1:2, the melting temperature was 80℃, dropping velocity was 30d/min, the condensate was silicon oil, the diameter of burette was 2 mm, the temperature of refrigerant was 10-15℃. The average weight of drop pill was 25mg. The quality standard of the drop pills was researched in this paper and the determination of polysaccharide from Radix Codonopsis was established.
