PartⅠEffect of mifepristone on the expression of CEACAM6 in chorionic villi of early pregnancyObjective:To investigate the effect of mifepristone on the expression of CEACAM6 in chorionic trophoblast cells of early pregnancy, therefore explore the potential anti-tumor mechanism of mifepristone. Methods:40 normal chorionic villi of early pregnancy were divided into two groups:medical abortion group (n=22) and surgical abortion group (n=18).The expression of CEACAM6 protein in chorionic villi were detected by immunohistochemistry, and the semi-quantity of CEACAM6 protein and mRNA were detected by western blot and RT-PCR. Results:The expression of CEACAM6 protein were found in cytotrophoblastic cells, syncytiotrophoblast cells and stroma cells. Both protein and mRNA of CEACAM6 in medical abortion group was significantly less than surgical group (p<0.05). Conclusion:Mifepristone could down-regulate the expression of CEACAM6 in chorionic villi of early pregnancy, that was one of the reasons leading to abortion. It also suggested that mifepristone should exert an anti-tumor effect through the same mechanism.PartⅡEffect of mifepristone on proliferation and expression of CEACAM6 of human ovarian cancer cell line SKOV3Objective:To evaluate the effect of mifepristone on proliferation and expression of CEACAM6 of human ovarian cancer cell line SKOV3 in vitro. Methods:Human ovarian cancer cell line SKOV3 was cultured with different concentrations (control group,1×10-7,1×10-6,1×10-5,5×10-5mol/L) of mifepristone(MIF) in vitro; MTT was used to examine the cell growth; and the semi-quantity of CEACAM6 mRNA and protein were detected by RT-PCR and western blot. Results:Proliferation of SKOV3 cultured with 1×10-7,1×10-6 mol/L of MIF was not significantly lower than the control group (p>0.05); 5×10-5mol/L of MIF were able to significantly suppress the proliferation of SKOV3 (p<0.01); The expressions of CEACAM6 mRNA and protein of SKOV3 in the control group were higher than that of 5×10-5mol/L of mifepristone group (p<0.05). Conclusion:Mifepristone could significantly suppress the proliferation of human ovarian cancer cell line SKOV3 in vitro. The action mechanisms may be associated with the suppression of expression of CEACAM6.
