Hepatocellular carcinoma (HCC) is one of common malignant tumor in our country, and its mortality is second only to gastric carcinoma and esophageal carcinoma, but first in Guangxi among all malignant tumors.Accounting to deaths in the word 45% of HCC each year.the mortality is about 11000 people in China.HCC metastasis is main reason leading to increased mortality. The molecular mechnism of HCC metastasis remained unclear. The research of HCC is concentrated on related gene cloning, cellular signal transduction pathway and regulation of cell cycle. Wnt signal pathway was a signal transduction pathway regulated by Wnt gene. It is demonstrated that Wnt signal pathway may not only play a critical role in the regulation of embryo development but also in the generation and progress of mang malignant tumors. Twist gene, possessing the featrue of oncogene, is an essential gene to tumor metastasis, and might play important role in inhibition of cellular apoptosis, invasion and metastasis of tumors.ObjectiveTo investigate the relationship between Twist gene and the Wnt signal pathway in MHCC97 Hepatoma cell line.MethodsWnt signal transduction pathway in MHCC97 Hepatoma cell line was activated by lithium chloride, a activator of Wnt pathway. Twist gene expression was detected by RT-PCR and fluorescence quantitative PCR.ResultsTwist gene expression in MHCC97 cell line activated by lithium chloride is higher than pre-activation (P<0.05).ConclusionIncreased expression of Twist gene might be induced by activated Wnt signal pathway in MHCC97 hepatocellular carcinoma cells with high metastatic potential, and it indicated that expression of Twist gene could be rugulated through Wnt signal pathway.
