Objective: To screen significant immune activity of 27 kinds of the analogues of unsaturated fatty acids Banlangen and further experimental study of skin graft.Methods: To observe the analogues on lymphocyte growth conditions of OD values detected by microplate reader, we had mouse lymphocytes reaction in vitro and mixed lymphocyte reaction of spleen. According to skin graft experi- ment, ear skin of allogeneic mouse was placed in wound bed on the back of the recipient. The skin from C57BL/6 mice was transplanted to BALB/c mice and the survival period of the grafts was assessed. Recipients were divided into four groups (physiological saline group, UFA231 group, P group, H group.). They were injected with physiological saline, UFA231, P, H (5mg·kg-1)once a day from one day to ten day after the operation.Result: Most of analogues had immunological effect. Seven kinds of the analo- gues with the more obvious effect of immune suppression and dose-dependent. Inhibition ratio of UFA231, K, P is more than 80 percent at high dose(250μg·ml-1),especially. UFA231, H, K showed a certain degree of immune suppression in the mixed lymphocyte,too. Effects of UFA231, P, H (5mg·kg-1) were not very obvious from inhibition of skin, in particular. H not only shortened the survival time of skin, but also promoted the skin off. The result was not consistent with mouse lymphocytes reaction in vitro. The reason needs to be further scientific researched. A series of experiments laid a solid foundation for mechanism of action of the mouse lymphocytes reaction in vitro.Conclusions: Most of analogues have immunological effect. Inhibition ratio of UFA231, K, P are more than 80 percent at high dose (250μg·ml-1). Effects of UFA231, P, H (5mg·kg-1) are not very obvious from inhibition of skin. Detailed mechanism needs further study.
