Objective:To isolate and identify unsaturated fatty acids from "Banlangen"-root of Isatis indigotica Fort.(Cruciferae),synthesize their analogues,and study their bioactivities of anticancer and hemolysis in vitro.Methods:Unsaturated fatty acids from "Banlangen"-root of Isatis indigotica Fort.were isolated and identified.Their analogues were synthesized from erucic acid in microscale organic laboratory and their chemical structures were elucidated with the spectroscopic methods,such as IR,1HNMR,13CNMR,MS and so on.In vitro,the growth and morphologic changes of the Tca8113 tongue carcinoma cells were observed in different concentrations of unsaturated fatty acids and their analogues.Their hemolytic effects on red blood cells were also detected.Results:Four compounds were isolated from "Banlangen" by column chromatography and two of them were identified as unsaturated fatty acids andβ-sitosterol,eleven compounds were synthesized and their structures were elucidated by spectroscopic analysis.The unsaturated fatty acids could kill cancer cells at high concentration (250μg/ml),and all compounds had hemolysis but the effects of the unsaturated fatty acids was more obvious.The application of microemulsion solved the poor water-solubility problem of compounds.Conclusion:The unsaturated fatty acids can kill cancer cells and all compounds have hemolytic effects.The carboxyl is the anticancer and hemolysis active group of the unsaturated fatty acids.
