objective:To observed the effect of different dose traditional Chinese medicine Achyranthes bidentata on rat neuronal apoptosis with severe traumatic brain injury, and to discuss the mechanism of brain injury treatment.Methods:78 rats were randomly divided into 6 groups:A:sham group, B: model group, C:naloxone group, D:Achyranthes high dose group, E: Achyranthes middle dose group, F:Achyranthes low dose group, each 13; Severe traumatic brain injury rat model was made by improved self-made Feeney's free-fall device, A group of just drilling the skull, not beating。A, B group with normal saline, C group received naloxone injection intraperitoneal injection, D, E, F respectively by high, medium and low dose to water decoction of Achyranthes bidentata; treatment for 1 week later, the brains were removed, paraffin sections, using TUNEL in situ detection of apoptotic neuronal damaged, Observed the effect of achyranthes bidentata on rat neural cell apoptosis with severe traumatic brain injury.Results:Compared with A (model group),the number of apoptotic cells of other groups was increased (P<0.05); and compared with B (model group): the number of apoptotic cells of D group (Achyranthes high dose group) significantly increased (P<0.05); E group (Achyranthes middle dose group) and F group (Achyranthes low dose group) apoptotic cells showed no significant difference (P> 0.05); The number of apoptotic cells naloxone group was no differences whenα= 0.05 as the test water standards。Conclusion:the number of apoptotic nerve cells D group (Achyranthes high dose group) traumatic brain injury rats was more than B group (model group), the difference was statistically significant; Compared with B (model group):the difference of the number of apoptotic nerve cells C group (naloxone group), E group (Achyranthes middle dose) and F group (Achyranthes low dose group) traumatic brain injury rats was was not statistically significant.
