Objective: In this study,spontaneously hypertensive rats(SHR)were taken as the research object.The antihypertensive effects of Angelica-Achyranthes bidentata Decoction and its fractions were analyzed and compared.The effects of Angelica-Achyranthes bidentata Decoction and its fractions on blood pressure were preliminarily explored through the changes of serum levels of ET-1,NO,VEGF and MMP-9.Method: In this experiment,D-101 macroporous resin was used to separate Angelica sinensis-Achyranthes bidentata decoction.Spontaneous hypertensive rats were randomly divided into eight groups.Except model group,the other seven groups were given valsartan,Angelica sinensis-Achyranthes bidentata decoction,pure water washing component,30%ethanol eluting component,50% ethanol eluting component,70% ethanol eluting component and 90% ethanol eluting component respectively for 4 weeks.Wistar rats of the same age were taken as the normal control group,while rats of the normal control group and the model group were given saline of the same volume and frequency simultaneously.The blood pressure of rats in each group was measured once a week before and after treatment by non-invasive blood pressure detection system.Four weeks after treatment,rats were killed under anesthesia and serum was collected.The levels of ET-1,NO,VEGF and MMP-9 in serum were detected by ELISA.Results SPSS 21.0 software was used for statistical analysis.Result:1.Macroporous resin was used to separate Angelica sinensis-Achyranthes bidentata decoction.The proportion of water-washing group was 68.03%,that of 30% ethanol-eluting group was2.53%,that of 50% ethanol-eluting group was 0.60%,that of 70% ethanol-eluting group was0.55%,and that of 90% ethanol-eluting group was 0.05%.The 30% ethanol elution group accounted for the highest proportion of the total ethanol elution components,accounting for67.73% of the total ethanol elution components.2.Comparing the therapeutic effects of each Chinese medicine treatment group on blood pressure of spontaneously hypertensive rats,Angelica sinensis-Achyranthes bidentata Decoction group and 30% ethanol elution group were better than other separation groups.3.The levels of ET-1 in serum of Angelica sinensis-Achyranthes bidentata Decoction group and 30% alcohol washing group were lower than those of model group,and the levels of NO were higher than those of model group(P < 0.05).4.The serum levels of vascular endothelial growth factor in Angelica-Achyranthes bidentata Decoction group,30% alcohol-washed group and water-washed group were significantly lower than those in model group(P < 0.05),and the levels of MMP-9 in Angelica-Achyranthes bidentata Decoction group and 30% alcohol-washed group were significantly lower than those in model group(P < 0.05).Conclusion:1.When Angelica sinensis-Achyranthes bidentata decoction was separated by macroporous adsorption resin,the antihypertensive effect of the 30% ethanol elution group was better than that of other separation groups,and the pharmacodynamic effect was closest to that of the original group.The 30% ethanol elution component might be the antihypertensive active part of the drug.2.Angelica-Achyranthes bidentata Decoction and its separated components may reduce blood pressure by regulating the levels of serum ET-1 and NO to improve vasodilation,regulating the levels of serum vascular endothelial growth factor and MMP-9 to improve vascular remodeling.Angelica-Achyranthes bidentata Decoction and 30% ethanol elution group had the most significant effect.
