In recent years, the research has shown that exogenous CNTF possess can promote repair of defect nerve, but the results are quite different. In this thesis, researches and experiments have been designed to further explore the effect of exogenous CNTF on the regeneration and motor functional resumption of nerve defect, which may lay the basic experimental foundation to solve clinical problems of functional recovery of injured nerves.Material and method The 40 adult Wistar rats were randomly divided into A, B, C, D groups of 10 rats. At first, The right hind limb sciatic nerve of rats in each group were cut off, to establish animal models of nerve defect. Then, the position of nerve defect were wrapped up with biodegradable membrane, to form nerve regenerative chamber. The different concentrations of CNTF were injected to the nerve regeneration chamber of rats in A, B, C groups. the same amount of saline were into rats of D group. On the 4, 8, 12 week after operation, the gait of rats were measured, and calculate sciatic functional index (SFI) by Bain formula. On the 12 week after operation, nerve muscle action potential (MAP) and motor nerve conduction velocity (MCV) of the damaged nerve were detected. Then cut nerve distal parts (regenerative nerve) which were wrapped with biodegradable membrane, and the target organ (gastrocnemius). Specimens were made into sections of LM stained by HE and osmium tetroxide, and TEM by conventional method. The morphological index of nerve regeneration and target organ change were detected, under light microscope and transmission electron microscopy. The data obtained were analysis by SPSS11.0 statistical package. Data are shown by mean±standard, analysis of variance was used to compare between the two groups, P values of "0.05 was considered statistically significant..Results are as following:1. On 8th , 12th week after operation, SFI from A, B, C group are more than one from group, and there is significant difference (P <0.05). SFI from C group is largest among A, B, C, D groups.2. On 12th week after operation, the MCV, MAP, wet weight and cross-sectional area survival rate of gastrocnemius from A, B, C group are higher than ones from D group, and there is significant difference (P <0.05). Among A, B, C, D groups, MCV from C group is the fastest, MAP from C group amplitude is the maximum.3. On 12th week after operation, despite average number of the regenerative myelinated nerve of the nerve distal part were less than ones of the autologous nerve proximal part. but there is no significant difference (P>0.05) on the average number of the regenerative myelinated nerve between the nerve distal part and autologous nerve proximal part in the A, B, C group. Compared with the proximal part of autologous nerve and nerve distal part of A, B, C group, the average number of the regenerative myelinated nerve of the in D group were less, and there are significant difference (P <0.05). The quantity of regenerative myelinated nerve in C group is the largest among A, B, C, D groups.4. The diameter, cross-sectional area of myelinated fiber and thickness of myelin sheath from nerve distal part were less than autologous nerve proximal part, and there is significant difference (P <0.05). but, the diameter, cross-sectional area of myelinated fiber and thickness of myelin sheath from nerve distal par in A, B, C group are larger than ones in D group, and there is a significant difference (P <0.05).The diameter, cross-sectional area of myelinated fiber and thickness of myelin sheath from regenerative myelinated nerve in C group is the largest among A, B, C, D groups.Based on the experiment, conclusion is shown as following::1. On 12th week after operation, although numbers of regenerative myelinated nerve close to the nerve proximal part, but there is significant difference on the diameter, cross-sectional area of myelinated fiber and thickness of myelin sheath.2.The certain concentration of exogenous CNTF in nerve regenerative chamber wrapped with biodegradable membrane can promote nerve structural regeneration and motor function resumption.3. Close relations exist between the promoting effect on regeneration of defect nerve and concentration of exogenous CNTF. In this experiment, the promoting effect on regeneration of defect nerve increase with augment of concentration of exogenous CNTF. its catalytic effect rendered with the CNTF was significantlyd (P <0.05) trend. 4. Biodegradable membrane in the repairing process of of peripheral defect nerve shown good results and application prospects.
