Periodontitis is a kind of infection-based diseases, periodontal support the organization, once lost, it will be difficult to restore its original structure and function. Therefore, the fundamental purpose of periodontal treatment is to control the occurrence and development of the disease, and recovery the destruction. Many studies showed that, PDLC is a major cellular components of periodontal membrane and is the precursor cells of periodontal tissue regeneration. In the periodontal tissue regeneration process, a certain number of healthy periodontal ligament cells are the basis for periodontal tissue regeneration and repair. Periodontal precursor cells to migrate and occupy the crown to the root surface is the key to the formation of periodontal tissue regeneration, the migration process is affected by many factors, there include cells in their proliferative activity and peptide growth factors on the regulation of cell growth and differentiation. To a certain extent, PDLC proliferation and migration determines the prognosis of treatment.Syndecan-4 is a transmembrane heparan sulfate protein and composed by a core protein and glycosaminoglycan (GAG). Present studies suggest that cell Syndecans region, especially Syndecan-4 region in cell to be directly involved in signal transduction. Some reports indicate that Syndecan-4 like growth factor,induced cell proliferation of media and migration. In addition, Syndecan-4 PDZ interaction region mutations, bFGF led to injury of endothelial cell migration and proliferation of failed or delayed. Syndecan-4 cytoplasmic tail region existence is necessary for the response of bFGF.FGFs have a wide range of biological effects on cell proliferation, differentiation, migration and animal tissue repair. FGFs play a role through the effects cell of specific surface receptors. FGFs have at least three types of receptors: tyrosine kinase receptor, cysteine-rich FGF receptor and heparin sulfate glycoprotein. Now basically confirmed the third receptors of FGF may be the FGF combination of heparin sulfate glycoprotein. Because of its close connection with the heparin, which is also known as heparin-bound FGF receptors. Since HSPGs can be at a lower level (nM level) combined with FGF, which is called low-affinity receptor. bFGF is one of the FGFs are heparin-binding peptides, distributed in a variety of organizations, with a variety of biological functions, bFGF is a kind of FGFs ,distribution in a variety of organizations, with a variety of biological functions and it is important to mitosis-promoting factor, but also morphogenesis and differentiation-inducing factor. In addition, cell surface heparin sulfate proteoglycan chains as the signal transduction mechanism of bFGF,biological effect of bFGF under the control of Syndecan-4, bFGF should first linked with heparan sulfate part of the extracellular domain of Syndecan-4,then combined with the high-affinity receptors play a p romoting cell proliferation, migration and differentiation.Methods: We gathered health premolars of young people because of the removal of the orthodontic Oral and Maxillofacial Surgery in the collection of Jilin University ,washed tooth with stroke-physiological saline solution, cleaned blood and bacteria at surface of the root. We scraped the periodontal ligament in root 2/3 use of subgingival curette, using tissue explant cultured human periodontal ligament cells in vitro. PDLC were stimulated by bFGF in different concentration and without bFGF as a negative control group, then gene expression of Syndecan-4 was assessed by Real-time fluorescent quantitative PCR . Results: At an early stage , with the increasing concentration of exogenous bFGF, Syndecan-4 expression was significantly increased. The promotion was weakest in 72h,1.0ng/ml, and the strongest in 24h,1.0ng/ml. Conclusion: This regulation of bFGF on Syndecan-4 in human PDLC ,the effect may be one of important factors which participate in the proliferation,migration of PDLC.In short, Periodontal tissue regeneration is a very complex process, the specific role of the migration and proliferation processes are not very clear. Combined with our previous experimental study,We explore the mechanisms of bFGF in periodontal regeneration through studies of effect of bFGF on Syndecan-4 gene expression in periodontal ligament cells and it could provide a t heoretical basis for periodontal regeneration. Study in roles of each growth factor in periodontal tissue regeneration and the combined effects of growth factors also need long time.
