ObjectivesTo detect the expression and the clinical significance of the Lewis y antigen and TGF-β1 (transforming growth factor betal) in ovarian epithelial tumors, then to evaluate the correlation between them.MethodsImmunohistochemical staining was used to detect the expression of Lewis y antigen and TGF-β1 in 60 cases of ovarian epithelial malignant tumors(including 20 cases of high differentiation 20 cases of middle differentiation and 20 cases of low differentiation),20 cases of borderline ovary tumor,20 cases of benign ovary tumor and 10 cases of normal ovarian tissues. The relationship between Lewis y antigen and TGF-β1, between them and biologic characteristics of ovarian carcinoma was analyzed.All specimens were stained immunohisto-chemically for Lewis y and TGF-β1 identified immunohistochemically with specific antibody. Observed slices under 400 double light microscope,photoed under digital camera, input fields random seleted into MetaMorph,converted staining intensity of cells to quantification index, calculated opital density. Immunofluorescence double labeling method was also used to detect the correlation between Lewis y antigen and TGF-β1.Results1.The positive rates of Lewis y antigen in ovarian epithelial cancer tissues was 88.33%, obviously higher than that of borderline ovary tumors (60.00%), (P< 0.05),benign ovary tumors (35.00%), (P< 0.01) and normal ovarian tissues(0), (P< 0.01), Its expression were not assoiatced with clinic parameter;2.The positive rates of TGF-β1 in ovarian epithelial cancer were 78.33%, obviously higher than that of benign ovary tumors (65.00%) (P< 0.05), and normal ovarian tissues(50%), (P< 0.05); 3.The positive rates of the TGF-β1 and Lewis y were not assoiatced with metastasis of lymph nodes and histological types, differentiation degree and clinic stage (P>0.05); Expressions of Lewis y antigen and TGF-β1 were significant positively associated with epithelial.Conclusions1.The expression of Lewis y antigen and TGF-β1were significantly increased in ovarian cancer, that they promoted the development of the ovarian epithelial cancer.2.Lewis y antigen and TGF-β1 were positive correlation. Altered expression of Lewis y antigen may cause the change of TGF-β1. They were associated with the process of carcinogenesis and development of ovarian carcinomas. Lewis y antigen is an important part of TGF-βreceptor, we speculated that Lewis y antigen increase the growth of ovarian cancer cell and invasion ability by promote TGF-β1 abnormal expression and by promoting angiogenesis, change in its signal transduction pathway.
