The Study About Expression And Significance Of CD4~+CD25~+T Cell In Different Liver Tumor

ObjectiveTo study the distribution of CD4+CD25+regulatory T cells (Treg) in normal liver tissue,the liver focal nodular hyperplasia(FNH),atypical adenomatous hyperplasia of the liver(AAH), hepatocellular carcinoma (HCC) and side of cancer. We will evaluate its the role in tumor occurrence and development.Methods90 cases were divided into 4 groups:10 specimens with normal liver tissue,10 specimens with the liver focal nodular hyperplasia,10 specimens with atypical adenomatous hyperplasia of the liver.60 specimens with hepatocellular carcinoma. The expression of treg cells were measured in 90 cases with double ELISA method of immunohistochemical staining.The first, blind measurement method is used to observe entire sections in the low magnification microscope.we randomly selected 5 high power field to count positive cell, seeking their average, Analysis of its expression and significance in hepatocellular carcinoma.Rusult1. Normal liver tissue, FNH group were not found in the expression of Treg cells. AAH group have the expression of a small number of Treg cells, an average of a single high power field 2.20±1.14; HCC group, there is the expression of Treg cells, a single high power field an average of 6.78±2.16. Average of treg cells in side of cancer as a single high power field is average of 5.19±1.87.2. Treg cells of HCC group was signifieantly higher than in AAH group, and the difference between the two groups was statistically significant (P=0.000). HCC group and the adjacent tissue between the two groups was statistically significant difference. (P=0.002).3. Treg cells in the organization of the distribution of HCC:the expression of Treg Cells was no significant difference between different age and sex and preoperative AFP value (P> 0.05); It is a relationship between treg cells number and the size of tumor, and the difference between the two groups was statistically significant (P=0.028);Treg cells of Enveloped group was signifieantly higher than in no capsule group, and the difference between the two groups was statistically significant (P=0.035).Comparison between normal and upper groups of Preoperative HBV-DNA was statistically significant difference.(P=0.007).Conclusion1. FNH and normal liver tissue have non-Treg cells, AAH and HCC tissuesand he adjacent tissue have the expression of Treg cells.2. The expression of Treg cells in hepatocellular carcinoma has nothing to do with age, sex and preoperative AFP value. It is a relationship between the expression of Treg cells and the tumor size. It is also a relationship between the expression of Treg cells and Enveloped and HBV-DNA value.3. The increase of Treg cells favor occurrence and development of the tumor. It is plays a negative regulatory role in the immune of the tumor. Remove or reduce the Treg cells of the liver micro-environment may increase the effect of tumor immunotherapy.
