ObjectiveTo investigate the expressions of human epidermal growth factor receptor-2 (HER-2) and matrix metalloproteinase-2 (MMP-2) in human colorectal carcinomas and the clinical significance.MethodsThe immunohistochemical SP method was used to detect the expressions of the two proteins in 64 cases of colorectal carcinoma and 15 cases of adenoma.12 cases of paracancerous tissues as the control groups. Expression of HER-2 and MMP-2 were examined in resected specimens of colorectal carcinomal patients and other specimens with SP immunohistochemical staining.ResultsThe expressions of the two proteins in 64 cases colorectal carcinoma and 15 cases of adenoma were higher than those 12 cases of paracancerous tissues., there was significant relationship among them (P< 0.05),whereas the expressions of the two proteins of the colorectal carcinoma group and adenoma the group have no significant relationship (P>0.05).In the cases of colorectal carcinoma,the expression rates of HER-2 and MMP-2 were not related to sex,age,degree of differentiation and invasion depth (P>0.05), whereas related to the Dukes stages, metastasis of lym phnode (P<0.05). Among Dukes staging the positive rates of HER-2 and MMP-2 in stages of C and D were higher than those in stages of A and B (P>0.05),the positive rate of HER-2 and MMP-2 in colorectal carcinoma with lymph node metastasis were higher than those without lymph node metastasis. The expression rate of HER-2 was positively correlated with the expression of MMP-2 in colorectal carcinoma(r=0.303, P<0.05).ConclusionsThe expressions of HER-2 and MMP-2 in paracancerous tissues,adenoma and colorectal carcinoma were moving on,they are were highly expressed in the colorectal cancer. HER-2 and MMP-2 may promote the metastasis and evolution of colorectal cancer. HER-2 and MMP-2 maybe operate cooperative effect with the evolution of the colorectal carcinoma.
