ObjectivePancreatic cancer is one of the common malignant tumor in digestive system, its highly malignant occult onset, the prognosis is poor. Analysis of pancreatic cancer development mechanism and its inherent molecular biology, in order to find effective means of diagnosis and treatment of pancreatic cancer research has become the focus of many years,This sturdy is to detect the express of connective tissue growth factor (CTGF) and hypoxia inducible factor-la (HIF-1α) in pancreatic carcinomas and investigate their respective relationship with clinicopathologic features and the correlation.MethodsCollect 42 cases of paraffin embedded pancreatic carcinomas.Among them,the ration of sex man and woman was 30:12;the age varied from 43 to 79 years old and the median age was 59.5;TNM stage;9 cases of I stage,20 cases ofⅡstage,11 cases of III stage and 2 cases of IV stage;Among them,16cases were well-differentiated, and 26 cases were moderately and poor-differentiated; 18cases had lymph node metastasis and 24 without. And 10 cases of normal pancreases tissues.Immonohistochemistry staining (SP method) was used to detect expression of CTGF and HIF-la in 42 cases of paraffin embeded pancreatic carcinomas and 10 cases of normal pancreases tissues.The relationship between CTGF,HIF-la expression and clinic parameters of patients was analyzed and the correlation between CTGF expression and HIF-la expression was also analyzed.Results1.CTGF is expressed mainly in endochylema or cytomembrane of tumor cell,also in interstitial cell, In pancreatic carcinomas,the positivity rate of CTGF was 73.8% which was significantly higher than 30% in normal pancrease tissues(p<0.05).2.HIF-la is expressed mainly in endochylema or nucleus of tumor cell, In pancreatic carcinomas,the positivity rate of HIF-la was 71.4% which was significantly higher than that in normal pancrease tissues(p<0.05).3.In pancreatic carcinomas, CTGF expression was nothing to do with age,sex,differentiation, lymph node metastasis and clinical stage.HIF-la expression was closely correlated with lymph node metastasis and clinical stage,rather than age,sex and differentiation.Other-Wise,the expression of CTGF was relevant to the expression of HIF-1 a.Conclusions1.In pancreatic carcinomas,the positivity rate of CTGF was significantly higher than that in normal pancrease tissues, but CTGF expression was nothing to do with age,sex,differentiation, lymph node metastasis and clinical stage.2.In pancreatic carcinomas,the positivity rate of HIF-1a was significantly higher than that in normal pancrease tissues, and HIF-1a expression was closely correlated with lymph node metastasis and clinical stage.3.The expression of CTGF was relevent to the expression of HIF-la..The abnormal expression level of CTGF and HIF-la in pancreatic carcinomas was relevent to tumor' growing and lymph node metastasis. Detection of CTGF and HIF-la expression might be helpful to predict prognosis of patients with Pancreatic cancer.
