ObjectiveThe objective of this study was to examinate the expression of CyclinDl and NF-κB protein in colorectal carcinoma and the differences and correlations between the expression of CyclinDl, NF-κB protein and colorectal carcinogenesis.MethodsClinical data and tisue microarry construction:Surgically resected specimens of 131 cases of colorectal carcinoma.76 cases were male and 55 females; aged 31 to 81 years, median age 55 years; pathology are adenocarcinoma, well differentiated in 22 cases,90 cases of differentiated, poorly differentiated 19 cases; violation of muscle in 21 cases, violations of pulp film in 110 cases; lymph node metastasis in 48 cases,83 cases without metastasis. TNM staging according to cancer staging manual (AJCC) sixth edition of the TNM staging system, specifically into:I in 14 cases, II of 67 cases, III of 45 cases, IV of 5 cases.Immunohistochemical (IHC) method and assessment:Tissue microarrys including colorectal carciomas were constructed using Microarrayer,4μm consecutive sections were cut. Known positive tissue pieces positive control, using PBS instead of primary antibody as negative control. CyclinDl and NF-κB protein was located in the cytoplasm. or nucleus showed clear and specific immune response in brown particles positive cells. Staining intensity (A) was classified as 0 (negative),1 (weak),2 (moderate) and 3 (strong). The percentage of positive cells (B) examined in 200 cells were divided into 0 (<5%),1 (5%-25%),2 (26%-50%),3 (51%-75%) and 4 (>75%). According to the product of A and B, the IHC result was classified as 0, negative (-); 1-4, weakly positive (+); 5-8, moderately positive (++) and 9-12, strongly positive (+++). ResultsThe results of IHC show that the positive rates of CyclinD1 and NF-κB protein were 82.4%and 85.5%respectively. In 131. cases of colorectal carcinoma, the expression of CyclinD1 and NF-κB protein was significantly positively correlated, rk=0.394, P<0.001. CyclinD1 and NF-κB had no relationship with gender, age, lymph node metastasis, pathological type, depth of invasion (P>0.05).NF-κB in the tumor tissue of rectal cancer in poorly differentiated carcinoma was significantly higher than in well-differentiated and moderately differentiated carcinoma, it is associated with tumor pathology (rk=-0.451, P=0.002).ConclusionsNFκB expression and on behalf of the malignant behavior of rectal cancer is closely related to the degree of differentiation, suggesting that the prognosis of rectal cancer may have important value.. Our results showed a positive correlation between CyclinD1 and NF-κB expression, indicating that they may participate in the development in colorectal cancer of a synergistic effect may exist, but the molecular mechanisms still requires further investigation.
