Soluble soybean polysaccharide (SPS) was degraded by hydrogen peroxide (H2O2) from average molecular weight of 115,200 to 10,200, and the content of carboxyl group (-COOH) was decreased from 13.8% to 11.8%. SPS degraded by this method has a high efficiency, reagent consumption, easy operation, at the same time the content of carboxyl group was decreased slightly, solubility increased greatly, also the biological activity enhanced.Modulation of soluble soybean polysaccharide (SPS) and degraded soluble soybean polysaccharide (degraded SPS) solution on formation of Calcium Oxalate Crystal were attempted via in vitro biomineralization method by means of X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM) and ICP. SPS not only inhibited calcium oxalate monohydrate (COM) crystals growth, but also induced formation of thermodynamic metastable calcium oxalate dihydrate (COD) crystals.As the mass concentration of SPS increased from 0 to 0.20 g/L, the percentage of COD increased from 0 to 67.3%, the soluble calcium ions increased from 48.7μmol/L to 198μmol/L, and the mole of precipitation of calcium oxalate crystals decreased from 1.917 mmol to 1.884 mmol. After degraded by hydrogen peroxide (H2O2), the inhibited ability of SPS on growth of calcium oxalate increaded markedly. When the concentration of degraded SPS was 0.20 g/L, it can induce formation of 91.3% COD crystals, and the soluble calcium ions increased to 308μmol/L, at the same time the gathering trend of calcium oxalate crystals reduced considerably, also the sharp of calcium oxalate changed from sharp, plate-like crystals to blunt, tetragonal bipyramidal crystals. As COD crystals might adhere weaker than COM crystals to the tubule cell surface in kidney sections, it will beneficial to prevention and cure of urinary stones.This paper investigated the difference of inhibitory effect of SPS and degraded SPS on growth of calcium oxalate crystals, we expected to provide new insight in drugs exploitation of urinary stone therapy and prevention of urolithiasis.
