ObjectiveThe purpose of this study was to establish an experimental model of beagle dogs pulpitis induced by lipopolysaccharide (LPS).The exposed pulps were capped with three different agents: baicalin sodium alginate gel combined with mineral trioxide aggregate (MTA), Dycal,MTA.The therapeutic effects of direct capping were assessed through immunohistochemistry at different time point. The result of this study will provide experimental evidences and theoretical basises for using baicalin sodium alginate gel combined with MTA to cure pulpitis.MethodsTwo healthy, SPF level beagle dogs(age:12~14 months) were randomly divided into two groups,which are 7 days group and 30 days group.Each beagle dog was established an experimental model of pulpitis induced by LPS. A therapy of pulp capping will be processed with baicalin sodium alginate gel combined with MTA. Dycal,MTA were used as pulp capping agents in control groups.The dogs were sacrificed at 7th day and 30th day after arteria carotis perfusion fixation. Taking out the experimental teeth to make the paraffin and thinest slices. Immunohistochemical stains were performed to identify the change of tumor necrosis factor-a(TNF-a)expression before and after pulp capping.Results1 At 7th day after pulp capping, the rate of TNF-a positive cells in baicalin sodium alginate gel combined with MTA group was lower than those in pulpitis, Dycal group and MTA group(P<0.05),higher than those in normal pulp(P<0.01).The rate of TNF-a positive cells in MTA group was lower than those in pulpitis (P<0.05).The rate of TNF-a positive cells in Dycal group was obviously higher than those in normal pulp(P<0.01).2 At 30th day after pulp capping, the rate of TNF-a positive cells in baicalin sodium alginate gel combined with MTA group was lower than those in pulpitis, Dycal group and MTA group(P<0.05). The rate of TNF-a positive cells in MTA group was higher than those in normal pulp (P<0.01), lower than those in pulpitis, Dycal group (P<0.05).The rate of TNF-a positive cells in Dycal group was higher than those in normal pulp (P<0.01), lower than those in pulpitis (P<0.01).Conclusions1 The method to establish an limited experimental model of beagle dog pulpitis induced by LPS was feasible.2 It had better effects using baicalin sodium alginate gel combined with MTA as pulp capping agents in pulptis than Dycal or MTA, with having advantage of reducing the expression of TNF-a.
