Objective:To investigate mechanisms of Metastasis-Associated gene 1(MTA1) promoting colorectal carcinoma lymphangiogenesis and metastasis.Methods:The expression of MTA1,HIF-la, and D2-40 marked lymphatic vessels were detected by immunohistochemical staining in eighty-one human colorectal carcinoma cases; Human colon carcinoma cell line LOVO was transfected with encoding human MTA1 and enhanced green fluorescent fusion protein(EGFP-MTA1) recombinant vector by liposome and established stably expressing MTA1 cell line.On the other hand,Down-Regulation of MTA1 expression using small interfering RNA in human colon carcinoma cell line SW480,Then detect the changes of the VEGF-C/D mRNA expression in LOVO and SW480 cells by RT-PCR after their transfected.Results:MTA1 and HIF-la expression has a positive correlation in 81 human colorectal carcinoma cases(P<0.05); The expression of MTA1 and HIF-la both correlated with Duke's stage, peritumoral lymphatic vessel density(LVD),lymph node metastasis (P<0.05), but did not correlate with the pathological grade and depth of invasion(P>0.05);In MTA1 overexpressioned LOVO cells the VEGF-C mRNA expression was increased(P<0.05),but in MTA1 down-regulated SW480 cells,the VEGF-D mRNA expression was decreased(P<0.05).Conclusion:In colorectal carcinoma,the overexpression of MTAl promoted peritumoral lymphangiogenesis and lymphatic vessel metastasis, MTA1 and HIF-la expression has a positive correlation;MTA1 may induce lymphangiogenesis by regulation of VEGF-C/D expression through and futher promote the lymphatic vessel metastasis in colorectal carcinoma.
